細(xì)胞因子在雌小鼠內(nèi)毒素攻擊中的變化及其對(duì)卵母細(xì)胞成熟影響機(jī)理的研究

【摘要】:研究背景和研究思路 內(nèi)毒素(endotoxin)是存在于革蘭氏陰性菌外膜中的大分子脂多糖(lipopolysaccharide,LPS)物質(zhì),是感染性休克的主要致病因子,而細(xì)胞因子如腫瘤壞死因子α(tumor necrosis factor-alpha)、表皮生長(zhǎng)因子(epidermal growth factor,EGF)、內(nèi)皮素-1(endothelin ET-1)、一氧化氮(Nitric oxide,NO)等在介導(dǎo)或保護(hù)LPS引起的病理生理學(xué)變化中有著非常重要的作用。故而,阻斷細(xì)胞因子引起的炎癥級(jí)聯(lián)反應(yīng),,在防治內(nèi)毒素攻擊中具有重要的意義,然而至今尚未尋找到有效的內(nèi)毒素拮抗劑。 雌性激素是存在于女性體內(nèi)的正常激素,包括雌激素(以雌二醇—estradiol,E2為代表)和孕激素(以孕酮—progesterone,P為代表)。E_2和P除對(duì)生殖系統(tǒng)有影響外,還對(duì)機(jī)體有重要的保護(hù)性作用。人們已經(jīng)知道,婦女更年期后,易發(fā)現(xiàn)骨質(zhì)疏松和骨折,心血管疾病的發(fā)病率也顯著升高,即雌性激素對(duì)運(yùn)動(dòng)系統(tǒng)和心血管系統(tǒng)具有保護(hù)作用,F(xiàn)在已經(jīng)知道,雌性激素對(duì)免疫系統(tǒng)、神經(jīng)系統(tǒng)、消化系統(tǒng)等均發(fā)揮調(diào)節(jié)和保護(hù)作用。更重要的是,最近有一系列實(shí)驗(yàn)證據(jù)證實(shí)雌性激素對(duì)燒傷、創(chuàng)傷、感染等傷害性刺激造成的免疫系統(tǒng)、神經(jīng)系統(tǒng)、心血管系統(tǒng)和消化系統(tǒng)等的功能紊亂和器質(zhì)性損傷具有保護(hù)作用。但是,雌性激素的水平隨生殖周期的變化而呈現(xiàn)周期性的變化,E_2含量于動(dòng)情前期逐漸升高,至動(dòng)情期達(dá)高峰,動(dòng)情間期最低;P含量于動(dòng)期后期達(dá)高峰,動(dòng)情間期最低。然而,在動(dòng)情周期的不同階段行內(nèi)毒素攻擊,對(duì)肝、肺等組織造成的病理?yè)p傷有無(wú)差異尚不清楚,其血漿和卵巢中內(nèi)源性
[Abstract]:Research background and research ideas Endotoxin (endotoxin) is a macromolecule lipopolysaccharide in the outer membrane of Gram-negative bacteria, which is the main pathogenic factor of septic shock. However, cytokines such as tumor necrosis factor-alpha, epidermal growth factor, endothelin et 1, nitric oxide (no) play an important role in mediating or protecting pathophysiological changes induced by LPS. Therefore, blocking the inflammatory cascade induced by cytokines plays an important role in the prevention and treatment of endotoxin attack. However, no effective endotoxin antagonist has been found so far. Estrogen is a normal hormone in women, including estrogen (estradiolol E2) and progesterone (progesterone P as the representative). Besides affecting the reproductive system, estradiol E 2 and P also play an important protective role in the body. It has been known that after menopause, osteoporosis and fracture are easy to be found, and the incidence of cardiovascular disease is also significantly increased, that is, estrogen has protective effect on the sports and cardiovascular system. It is now known that estrogen regulates and protects the immune system, nervous system, digestive system, etc. More importantly, there has been a recent series of experimental evidence to prove the effects of estrogen on the immune system, nervous system, and other noxious stimuli such as burns, injuries, infections, etc. The dysfunction of cardiovascular system and digestive system and organic damage have protective effect. However, the level of estradiol showed a periodic change with the change of reproductive cycle. The content of E _ 2 increased gradually in the early estrus and reached its peak in estrus, the lowest content of P in estrus reached the peak in the latter stage of estrus and the lowest in estrus. However, it is not clear whether endotoxin attack at different stages of estrous cycle can cause pathological damage to liver, lung and other tissues, and endogenous in plasma and ovary.
