【摘要】：目的: 以一系列同一親本來(lái)源對(duì)氟康唑敏感性不同的白念珠菌為對(duì)象,研究其菌絲相與酵母相ERG11基因序列及分泌性酸性蛋白酶活性之間的差異,從而為白念珠菌病原真菌學(xué)和臨床檢驗(yàn)學(xué)中,究竟是選擇白念珠菌的菌絲相還是酵母相為研究對(duì)象提供理論依據(jù)。 方法: 對(duì)12株分離自同一HIV陽(yáng)性者的白念珠菌分別進(jìn)行菌絲相與酵母相培養(yǎng)后,抽提基因組DNA,PCR擴(kuò)增ERG11基因(包括部分上游和下游非編碼區(qū)),PCR產(chǎn)物用Acc Ⅰ和Mun Ⅰ消化,消化產(chǎn)物進(jìn)行瓊脂糖凝膠電泳(PCR-RFLP);選擇其中5株白念珠菌,用下游引物對(duì)部分ERG11基因進(jìn)行測(cè)序,比較兩相細(xì)胞ERG11基因序列差異;用牛血清白蛋白培養(yǎng)基測(cè)定8株白念珠菌菌絲相與酵母相分泌性酸性蛋白酶的活性。 結(jié)果: 1.CA-7、CA-14,CA-16和CA-17的菌絲相與酵母相ERG11基因酶切圖譜存在差異; 2.CA-7菌絲相與酵母相ERG11基因第1547位點(diǎn)、1587位點(diǎn)和1617位點(diǎn)的堿基存在差異;
[Abstract]:Objective: A series of Candida albicans with different sensitivity to fluconazole from the same parent were used to study the difference of ERG11 gene sequence and secretory acid protease activity between hyphae and yeast. It provides theoretical basis for the selection of mycelial phase or yeast phase of Candida albicans in mycology and clinical laboratory.
