【摘要】:前言 可注射性水凝膠是國際上近幾年興起的一種相對新穎的軟骨組織工程材料,它提供了一類新的細(xì)胞固定機制,可注射性材料用于軟骨缺損修復(fù)的優(yōu)勢在于材料與修復(fù)細(xì)胞之間嵌合連接好、塑形容易,細(xì)胞和生長因子能與材料充分均勻混合,且能通過關(guān)節(jié)鏡或其它微創(chuàng)方式操作,安全、有效、費用低。本實驗利用可注射性水凝膠——幾丁質(zhì)作為骨髓基質(zhì)干細(xì)胞載體,在體外進(jìn)行聯(lián)合培養(yǎng),為構(gòu)建組織工程軟骨組織提供實驗依據(jù)。 材料與方法 1 實驗動物 成年SD大鼠10只,雄鼠體重120—140g,由中國醫(yī)科大學(xué)實驗動物中心提供。 2 主要試劑和儀器 幾丁質(zhì)(SIGMA公司),DMEM(GIBCO公司產(chǎn)品),新生牛血清(GIBCO公司產(chǎn)品),percoll梯度分離液(PHARMACIA公司產(chǎn)品密度:1.073g/m),胰島素樣生長因子IGF-Ⅰ、轉(zhuǎn)化生長因子TGF-β_1(Sigma公司)、維生素C、地塞米松,CO_2培養(yǎng)箱,倒置顯微鏡。 3 幾丁質(zhì)凝膠的制備 在10℃下,將幾丁質(zhì)(SIGMA公司)10g放入研缽中,加丙酮40ml研磨10min;邊研磨邊緩緩加入冷濃HC1400ml,充分研磨,使呈糊狀。于4℃放置24h,之后用玻璃棉過濾,將濾液緩緩加入到盛有2000ml體積分?jǐn)?shù)50%乙醇的燒杯中,邊加邊劇烈攪拌,然后靜置,待膠態(tài)幾丁質(zhì)析出后去除清液,收
