脂肪干細(xì)胞體外分離培養(yǎng)鑒定及免疫學(xué)性質(zhì)

【摘要】:背景:脂肪干細(xì)胞由于具備易于獲取等特點(diǎn),在組織工程中比骨髓間充質(zhì)干細(xì)胞具有更加廣闊的應(yīng)用前景,并因此成為當(dāng)今的研究熱點(diǎn)之一。目的:觀察脂肪干細(xì)胞的生長規(guī)律,鑒定其干細(xì)胞特性和免疫學(xué)性質(zhì)。方法:自整形外科行抽脂的3名健康志愿者身上獲取皮下脂肪組織及外周血,志愿者年齡20-30歲,均為女性。用Ⅰ型膠原酶消化脂肪組織獲得脂肪干細(xì)胞,在體外原代培養(yǎng)及傳代培養(yǎng),觀察其體外增殖能力等生物學(xué)特性;通過流式細(xì)胞術(shù)檢測人脂肪干細(xì)胞分化群表面抗原和主要組織相容性抗原的表達(dá)。并將異體脂肪干細(xì)胞和淋巴細(xì)胞共培養(yǎng),觀察脂肪干細(xì)胞是否可以刺激后者增殖。結(jié)果與結(jié)論:所獲脂肪干細(xì)胞具有很強(qiáng)的體外增殖能力,其生長曲線呈"S"形;第3代脂肪干細(xì)胞表面CD29、CD44、CD90以及主要組織相容性抗原Ⅰ呈陽性表達(dá),CD31、CD45、CD34及主要組織相容性抗原Ⅱ呈陰性表達(dá);未發(fā)現(xiàn)異體脂肪干細(xì)胞刺激淋巴細(xì)胞增殖。提示所獲細(xì)胞確為脂肪干細(xì)胞,且具有較弱的排斥反應(yīng),有可能成為細(xì)胞治療或組織工程的同種異體細(xì)胞來源。
[Abstract]:Background: adipose stem cells have a wider application prospect than bone marrow mesenchymal stem cells in tissue engineering due to their characteristics of easy to obtain and therefore become one of the research hotspots. Objective: to observe the growth of adipose stem cells and identify their stem cell characteristics and immunological properties. Methods: subcutaneous adipose tissue and peripheral blood were obtained from 3 healthy volunteers aged 20 to 30 years. Adipose stem cells were obtained by digesting adipose tissue with type I collagenase. The expression of differentiation surface antigen and major histocompatibility antigen of human adipose stem cells were detected by flow cytometry. The allogeneic adipose stem cells and lymphocytes were co-cultured to observe whether adipose stem cells could stimulate the proliferation of the latter. Results and conclusion: the adipose stem cells had strong proliferative ability in vitro, and the growth curve was "S" shape.
