【摘要】: Rh血型系統(tǒng)是臨床輸血醫(yī)學(xué)中僅次于ABO血型系統(tǒng)的輸血因子,在人類血型系統(tǒng)中最為復(fù)雜的當(dāng)屬Rh血型系統(tǒng),它能導(dǎo)致溶血性輸血反應(yīng)和新生兒溶血病[1]。在臨床輸血工作中,對Rh血型的分型使用血清學(xué)方法,根據(jù)紅細胞上D抗原的有無,分為Rh陰性和Rh陽性。據(jù)報導(dǎo)Rh陽性在我國漢族及大多數(shù)民族中約占99.6%,個別少數(shù)民族為90%。近年來,隨著分子生物學(xué)研究的不斷深入,對Rh血型基因的研究廣泛開展,其中研究最多的是序列特異性聚合酶鏈?zhǔn)椒磻?yīng)(PCR-SSP),即根據(jù)RH基因的特異性序列,通過PCR擴增出特異性產(chǎn)物,研究Rh血型基因的多態(tài)性。本實驗采用PCR-SSP法,通過對Rh血型基因特異性序列的擴增,分析RH基因多態(tài)性,旨在建立適合Rh血型的基因定型方法,為應(yīng)用于臨床奠定基礎(chǔ)。
[Abstract]:Rh blood group system is the second only blood transfusion factor in clinical transfusion medicine after ABO blood group system. The most complex blood group system in human blood group system is Rh blood group system, which can cause hemolytic transfusion reaction and neonatal hemolytic disease [1]. In clinical blood transfusion, serological method was used to classify Rh blood group into Rh negative and Rh positive according to the presence or absence of D antigen on erythrocytes. Rh positive is reported to be about 99.6% of the Han and most ethnic groups in China, and 90% for individual minorities. In recent years, with the development of molecular biology, the research of Rh blood group gene has been carried out extensively, among which the sequence specific polymerase chain reaction (PCR-SSPN) is the most widely studied, that is, according to the specific sequence of RH gene, The specific products were amplified by PCR to study the polymorphism of Rh blood group gene. In this study, PCR-SSP method was used to amplify the specific sequence of Rh blood group gene and to analyze the polymorphism of RH gene. The aim of this study was to establish a gene typing method suitable for Rh blood group and to lay a foundation for clinical application.
