【摘要】：目的 本文在食餌法高脂血癥動物模型的基礎(chǔ)上建立痰阻血脈大鼠模型，通過觀察大鼠血漿血脂的變化來評價模型。通過觀察主動脈內(nèi)皮巨噬細胞與粘附分子的表達及巨噬細胞的形態(tài)，來探討痰邪致病的細胞和分子機制。同時，觀察固本化痰通脈方對模型大鼠的影響，，以反證模型成功與否，以及對痰證的治療作用，為臨床治療痰證提供一定依據(jù)。 方法 7周齡健康雄性Wistar大鼠50只，200±20g，隨機分為5組：正常組、模型組、固本化痰通脈方低劑量組(簡稱固低組)、固本化痰通脈方高劑量組(簡稱固高組)、西藥組。 觀察大鼠的一般情況，包括飲食情況、活動度、毛發(fā)光澤度等變化；處死后觀察各組大鼠血漿中TC、TG、HDL、LDL的變化；采用免疫組織化學(xué)方法檢測主動脈內(nèi)皮MΦ、ICAM-1、VCAM-1、P-selectin、E-selectin的表達，并經(jīng)圖像分析定量；運用透射電鏡觀察主動脈內(nèi)皮巨噬細胞的超微結(jié)構(gòu)。 結(jié)果 模型組大鼠普遍出現(xiàn)納差、少動、拱背嗜睡、毛色發(fā)黃、缺乏光澤及掉毛嚴重等表現(xiàn)。正常組大鼠活潑好動、毛色光亮、食欲旺盛。 模型組血脂水平與正常組相比具有顯著性差異(P＜0.01)；經(jīng)治療后，TC、TG、LDL-c降低，HDL-c升高；與模型組相比，固低組TG、LDL-c降低，HDL-c升高有差異(P＜0.05)，TC降低有顯著性差異(P＜0.01)；固高組和西藥組與之均有顯著性差異(P＜0.01)。與西藥組相比，固高組降TG有差異(P＜0.05)，其余無顯著性差異；固低組TC、HDL-c、LDL-c具顯著性差異(P＜0.01)，TG無顯著性差異。與固低組比，固高組TC、LDL-c有差異(P＜0.05)，TG、HDL-c有顯著性差異(P＜0.01)。 免疫組化結(jié)果顯示，正常組幾乎沒有巨噬細胞表達，模型組與之相比有顯著性差異(P＜0.01)，固低組與西藥組無顯著性差異(P＞0.05)，固高組與二者有顯著性差異(P＜0.01)；模型組ICAM-1、VCAM-1、P-selectin、E-selectin的表達升高，與正常組相比有顯著性差異(P＜0.01)，經(jīng)治療后表達降低，與固高組相比有顯著性差異(P＜0.01)。 超微結(jié)構(gòu)結(jié)果顯示模型組血管內(nèi)皮下內(nèi)膜水腫、增寬，甚至間斷不完整；血中單核細胞及淋巴細胞向內(nèi)彈力板浸潤，可見較多巨噬細胞，其胞漿內(nèi)含大量各級溶酶體及吞噬小泡，內(nèi)質(zhì)網(wǎng)及核糖體增多，胞膜外出現(xiàn)偽足；內(nèi)皮細胞形態(tài)發(fā)生改變，可見線粒體水腫，胞漿不均勻；中藥治療后，上述病理損害明顯減輕。
[Abstract]:Purpose Based on the animal model of hyperlipidemia with prey method, the rat model of phlegm blocking blood vessel was established, and the model was evaluated by observing the changes of plasma lipids in rats. The expression of endothelial macrophages and adhesion molecules and the morphology of macrophages were observed to explore the cellular and molecular mechanisms of phlegm pathogen. At the same time, to observe the effect of Guben Huatan Tongmai recipe on the model rats, to reverse the success of the model, as well as the treatment of phlegm syndrome, to provide a certain basis for clinical treatment of phlegm syndrome. Method A total of 50 healthy male Wistar rats aged 7 weeks were randomly divided into 5 groups: normal group, model group, Guben Huatan Tongmai prescription low dose group, Guben Huatan Tongmai prescription high dose group (referred to as Gugao group, western medicine group). The changes of diet, activity, hair glossiness and so on were observed, the changes of plasma TCU TGG HD-LDL and the expression of P-selectin in aorta endothelium M 桅 ICAM-1 VCAM-1 and P-selectin were detected by immunohistochemical method. The ultrastructure of aortic endothelial macrophages was observed by transmission electron microscope. Result In the model group, anorexia, motility, sleepiness, yellowish coat color, lack of luster and severe hair loss were found. Normal group rats were lively and active, bright coat color and strong appetite. There was significant difference in blood lipid level between the model group and the normal group (P < 0.01). There was significant difference between the two groups (P < 0.05) in decreasing HDL-c and the difference between the two groups (P < 0.01), and there was a significant difference between the two groups (P < 0.01). Compared with the western medicine group, there was significant difference in TG reduction between the Gugao group and the western medicine group (P < 0.05), but there was no significant difference in TG between the two groups (P < 0.01), but there was no significant difference between the two groups (P < 0.01). There was significant difference (P < 0.05) in LDL-c of TCU and HDL-c between the two groups (P < 0.01). The results of immunohistochemistry showed that there was almost no macrophage expression in normal group, there was significant difference between model group and model group (P < 0.01), there was no significant difference between Gu-low group and western medicine group (P > 0.05), there was significant difference between Gugao group and both groups (P < 0.01), and the expression of ICAM-1VCAM-1P-selectinin E-selectin was increased in model group. Compared with the normal group, there was a significant difference (P < 0.01), the expression decreased after treatment, and there was significant difference (P < 0.01) compared with the Gugao group. The ultrastructural results showed that the intima of vascular endothelium in the model group was edematous, enlarged, and even discontinuous incomplete, the monocytes and lymphocytes in the blood infiltrated into the elastic plate, and more macrophages were observed. Its cytoplasm contains a large number of lysosomes and phagocytic vesicles, the endoplasmic reticulum and ribosome increase, and pseudopodia appear outside the cell membrane. The above pathological lesions were obviously alleviated.
