【摘要】：將原發(fā)性肝細(xì)胞肝癌7721 細(xì)胞注入小鼠體內(nèi)進(jìn)行免疫,再將免疫小鼠的脾細(xì)胞同小鼠骨髓瘤653 細(xì)胞融合,經(jīng)過篩選和克隆,獲得了一種新的鼠源單克隆抗體Hepama-2,其亞型為IgG2b。病理組織學(xué)分析表明這種單克隆抗體所識(shí)別的抗原決定簇特異地分布在人體原發(fā)性肝癌的細(xì)胞表面上。本文除了報(bào)告該抗體的一般生物學(xué)和生物化學(xué)特性外,還克隆了它的可變區(qū)基因,分析了可變區(qū)的基因和蛋白順序,為進(jìn)一步的抗體人源化工作做好了準(zhǔn)備,此外,我們用western-blot 法證實(shí)該單抗所對(duì)應(yīng)的抗原分子量為85KD,又用TFMS 法證實(shí)該抗原分子屬糖蛋白。
[Abstract]:The 7721 cells of primary hepatocellular carcinoma were injected into the mice to be immunized, and then the spleen cells of the mice were fused with the 653 cells of the mice myeloma. After screening and cloning, a new murine monoclonal antibody Hepama-2 was obtained. The subtype was determined by the IgG2b. pathological analysis of the antigen identified by the monoclonal antibody. In addition to the general biological and biochemical characteristics of the antibody, this paper also cloned its variable region gene, analyzed the gene and protein sequence of the variable region, and prepared for further antibody humanization. In addition, we confirmed it by Western-blot. The molecular weight of the McAb is 85KD, and TFMS is used to confirm that the antigen is glycoprotein.
