自體血漿和慶大霉素對細胞因子誘導(dǎo)的殺傷細胞免疫表型和活性的影響

【摘要】:目的探索自體血漿添加量和慶大霉素對培養(yǎng)的細胞因子誘導(dǎo)的殺傷細胞(CIK細胞)免疫表型和活性的影響。方法采用流式細胞術(shù)分析自體血漿和慶大霉素對CIK細胞中不同亞型免疫細胞比例的影響,細胞計數(shù)、MTT法檢測處理后的CIK細胞對HeLa細胞的細胞毒性差異。結(jié)果自體血漿實驗組與對照組之間不存在顯著性差異,而慶大霉素實驗組與對照組相比,CD3+細胞、CD56+細胞、CD3+CD8+細胞和CD3+CD56+細胞的數(shù)量均下降,差異有統(tǒng)計學(xué)意義(P0.05),下降幅度分別為1.4、1.7、1.4和2.5倍。進一步研究發(fā)現(xiàn),慶大霉素實驗組與對照組的CIK細胞對HeLa細胞的細胞毒性分別為43.0%和90.0%,兩者存在顯著性差異(P0.05)。結(jié)論自體血漿對CIK細胞不同亞型比例無明顯影響,慶大霉素可引起CD3+細胞、CD56+細胞、CD3+CD8+細胞和CD3+CD56+細胞減少,有一定細胞毒性。
[Abstract]:Objective to investigate the effects of autogenous plasma addition and gentamicin on the immunophenotype and activity of cultured cytokine-induced killer cells (CIK cells). Methods the effects of autologous plasma and gentamicin on the proportion of different subtypes of immune cells in CIK cells were analyzed by flow cytometry. The cytotoxicity of treated CIK cells to HeLa cells was detected by cell count assay. Results there was no significant difference between the autologous plasma test group and the control group, but the number of CD56 CD8 cells and CD3 CD56 cells in Gentamicin group was decreased compared with the control group. The difference was statistically significant (P 0.05), with a decrease of 1.4 and 2.5 times, respectively. Further studies showed that the cytotoxicity of gentamicin CIK cells to HeLa cells was 43.0% and 90.0%, respectively, and there was a significant difference between the two groups (P 0.05). Conclusion there is no significant effect of autogenous plasma on the proportion of different subtypes of CIK cells. Gentamicin can induce the decrease of CD3 CD8 cells and CD3 CD56 cells in CD3 cells.
