神經(jīng)干細胞在大鼠脊髓內(nèi)存活及遷移的實驗研究

【摘要】: 神經(jīng)干細胞(Neural stem cells, NSCs)是廣泛存在于中樞神經(jīng)系統(tǒng)內(nèi)未分化的細胞,它具有自我增殖及多向分化的潛能,被移植回中樞神經(jīng)系統(tǒng)后,能進一步遷移、分化,與宿主神經(jīng)元整合并發(fā)揮神經(jīng)功能。NSCs體內(nèi)移植為神經(jīng)系統(tǒng)損傷修復(fù)和退行性病變的治療帶來新的希望,從而成為當今神經(jīng)科學(xué)研究的熱點之一。 本實驗采用無血清培養(yǎng)技術(shù)培養(yǎng)NSCs,并利用神經(jīng)干細胞的標記物對其進行鑒定,應(yīng)用神經(jīng)元、神經(jīng)膠質(zhì)細胞、少突膠質(zhì)細胞特異性標志物NSE、GFAP、O4對自然分化的NSCs進行免疫細胞化學(xué)染色,證明其多向分化潛能,同時用免疫組織化學(xué)染色技術(shù)觀察了移植至大鼠脊髓內(nèi)的NSCs存活及遷移情況。 結(jié)果顯示:應(yīng)用DMEM-F12培養(yǎng)基,內(nèi)添加B27、EGF及bFGF進行NSCs的培養(yǎng),獲得的細胞球呈Nestin及Musashi染色陽性。細胞在添加10%胎牛血清自然分化后,呈NSE、GFAP、O4染色陽性。NSCs在大鼠體內(nèi)移植2周BrdU免疫組化染色結(jié)果顯示:在移植的腦脊膜處可見陽性細胞,散在分布脊髓白質(zhì)內(nèi),脊髓中央管周圍及灰質(zhì)內(nèi)亦可見陽性細胞。 提示:本實驗成功獲得了神經(jīng)干細胞,獲得的神經(jīng)干細胞可以向神經(jīng)元、神經(jīng)膠質(zhì)細胞、少突膠質(zhì)細胞分化。將NSCs移植后入大鼠脊髓內(nèi),NSCs可以存活并向周圍遷移。
[Abstract]:Neural stem cells (NSCs) are undifferentiated cells in the central nervous system (CNS), which have the potential of self-proliferation and multi-differentiation, and can migrate and differentiate further after being transplanted back to the CNS. Integration with host neurons and transplantation of NSCs in vivo bring new hope for the repair of nervous system injury and the treatment of degenerative diseases, and thus become one of the hotspots in neuroscience research. In this experiment, NSCs were cultured by serum-free culture, and the neural stem cell markers were used to identify NSCs. Neurons and glial cells were used to identify NSCs. Oligodendrocyte specific marker NSE-GFAPO4 was used for immunocytochemical staining of naturally differentiated NSCs to prove its multidirectional differentiation potential. The survival and migration of NSCs transplanted into the spinal cord of rats were observed by immunohistochemical staining. The results showed that NSCs was cultured on DMEM-F12 medium supplemented with B27 EGF and bFGF, and the cells were positive for Nestin and Musashi staining. After adding 10% fetal bovine serum to natural differentiation, the cells showed NSE-GFAPO4 positive staining. The results of BrdU immunohistochemical staining showed that positive cells could be seen in the transplanted meninges and scattered in the white matter of spinal cord. Positive cells were also seen around the central canal of the spinal cord and in the gray matter. The results showed that the neural stem cells were successfully obtained, and the neural stem cells could differentiate into neurons, glial cells and oligodendrocytes. NSCs transplanted into rat spinal cord can survive and migrate around.
