鼠防御素cryptdin 4在小鼠內(nèi)臟分布及其腸炎模型中表達(dá)規(guī)律

【摘要】: 防御素是小分子陽離子抗菌肽,他們既是先天性免疫的效應(yīng)因子,也參與獲得性免疫的調(diào)節(jié)。目前對于哺乳動物抗菌肽基因的作用及其機制還不清楚。本研究通過檢測cryptdin 4在小鼠體內(nèi)的分布情況及其在腸炎模型中小鼠回腸段cryptdin4基因表達(dá)量水平的變化,為研究cryptdin 4作用機制提供理論基礎(chǔ)。 我們選擇了對小鼠回腸、氣管、輸精管、輸卵管四個位置進行免疫組化檢測。檢測結(jié)果發(fā)現(xiàn)只有在回腸腸腺基底部的潘氏細(xì)胞處有陽性結(jié)果,其它三處皆為陰性結(jié)果,提示鼠cryp 4在小鼠回腸段有所表達(dá)。 本實驗根據(jù)GeneBank中公布的cryp 4和小鼠GAPDH序列設(shè)計了兩對特異性引物,應(yīng)用熒光定量PCR技術(shù)對腸炎模型狀態(tài)小鼠回腸段cryp 4表達(dá)水平進行檢測。在灌藥的第24h時回腸內(nèi)cryp 4表達(dá)量升高,隨著炎癥的發(fā)展表達(dá)量降低;在第4天時表達(dá)量降到最低,隨后略有回升,但仍然低于正常水平。 本實驗的結(jié)論可以為進一步研究小鼠cryp 4參與機體免疫的作用及其機制提供理論基礎(chǔ)。
[Abstract]:Defensins are small cationic antimicrobial peptides, which are not only the effector of innate immunity, but also involved in the regulation of acquired immunity. The role of antimicrobial peptide gene in mammals and its mechanism are unclear. In this study, the distribution of cryptdin _ 4 in mice and the changes of cryptdin4 gene expression in ileum of mice were detected in order to provide a theoretical basis for studying the mechanism of cryptdin _ 4. Four locations of mouse ileum, trachea, vas deferens and fallopian tubes were detected by immunohistochemistry. The results showed that the positive results were found only in the Pans cells at the base of the ileum and the other three were negative. The results suggested that mouse cryp 4 was expressed in the ileum of mice. Two pairs of specific primers were designed according to the cryp _ 4 and mouse GAPDH sequences published in GeneBank. The expression of cryp _ 4 in ileum of mice with enteritis was detected by fluorescence quantitative PCR technique. The expression of cryp _ 4 in ileum increased at 24 h after administration, decreased with the development of inflammation, reached the lowest level on day 4, then increased slightly, but was still below the normal level. The conclusion of this study may provide a theoretical basis for further study on the role of cryp _ 4 in the immune function of mice and its mechanism.
