某院鮑曼不動(dòng)桿菌流行克隆株的分子特征

【摘要】:目的了解臨床分離鮑曼不動(dòng)桿菌的主要流行克隆株的變化趨勢(shì),揭示其分子流行的主要特征。方法篩選解放軍253醫(yī)院2010年6月—2012年12月臨床分離的74株不重復(fù)鮑曼不動(dòng)桿菌作為實(shí)驗(yàn)菌株,應(yīng)用基于序列分型的多重聚合酶鏈反應(yīng)(PCR)方法鑒定鮑曼不動(dòng)桿菌的主要流行克隆譜系;利用限制性?xún)?nèi)切酶ApaI對(duì)實(shí)驗(yàn)菌株基因組DNA進(jìn)行酶切和脈沖場(chǎng)凝膠電泳(PFGE)分析,并利用BioNumerics軟件進(jìn)行聚類(lèi)分析;應(yīng)用PCR及測(cè)序方法檢測(cè)10種耐藥相關(guān)基因(blaOXA-51-like、blaOXA-23-like、blaOXA-24-like、blaOXA-58-like、blaTEM、blaampC、armA、ISAba1、intI 1和intI 2)的攜帶情況。結(jié)果本組實(shí)驗(yàn)菌株克隆譜系以Group 1序列型為主,所占比例高達(dá)77.0%;PFGE帶型表現(xiàn)出多樣性,可大致分為13種圖譜,其中主要型別為A型(35株);耐藥相關(guān)基因blaOXA-51-like、blaOXA-23-like、blaTEM、blaampC、armA、ISAba1和intI 1的陽(yáng)性率分別為100.0%、54.1%、86.5%、66.2%、89.2%、90.5%和85.1%,未檢出blaOXA-24-like、blaOXA-58-like和intI 2。結(jié)論該院臨床分離的鮑曼不動(dòng)桿菌存在主要流行克隆株,與歐洲克隆譜系Ⅱ具有高度的同源性;多重耐藥相關(guān)基因的攜帶率較高;不同的序列型或PFGE型別中存在同一耐藥基因交叉分布的現(xiàn)象,說(shuō)明多種耐藥相關(guān)基因在鮑曼不動(dòng)桿菌中發(fā)生著水平轉(zhuǎn)移。
[Abstract]:Objective to investigate the variation trend of the main prevalent clones of Acinetobacter baumannii and reveal the main characteristics of the molecular prevalence of Acinetobacter baumannii.Methods 74 clinical isolates of Acinetobacter baumannii isolated from 253 Hospital of PLA from June 2010 to December 2012 were selected as experimental strains.The main prevalent clone lineages of Acinetobacter baumannii were identified by multiplex polymerase chain reaction (PCR) based on sequence typing, and the genomic DNA was analyzed by restriction endonuclease (ApaI) and pulsed field gel electrophoresis (PFGE).BioNumerics software was used to cluster analysis, and PCR and sequencing methods were used to detect the carrying of 10 genes related to drug resistance, such as blaOXA-51-like blaOXA-23-like blaOXA-like blaOXA-58-like blaOXA-58-like bla-TEMampCarmCarmCarmA, ISAba1 (intI1) and intI _ (2).Results the clone lineage of the experimental strains was dominated by Group 1 sequence type, the proportion of which was as high as 77.0% and showed diversity, which could be divided into 13 kinds of maps.Conclusion the clinical isolates of Acinetobacter baumannii have a high homology with European Clone 鈪,
