活化蛋白C對(duì)大鼠急性胃粘膜損傷保護(hù)治療作用的實(shí)驗(yàn)研究

【摘要】:目的探討活化蛋白C(APC)對(duì)急性胃粘膜損傷的保護(hù)治療作用。 材料及方法大鼠48只,隨機(jī)分為八組。A1、A2組為正常組,B1(WRS2h)、B2(WRS3.5h)組為模型組,C1(WRS2h)、C2(WRS3.5h)組為rhAPC治療組,D1(WRS2h)、D2(WRS3.5h)組為生理鹽水對(duì)照組。復(fù)制浸水束縛應(yīng)激(WRS)模型并計(jì)算潰瘍指數(shù)(UI)。C1、C2組在WRS開始前半小時(shí)通過(guò)鼠尾靜脈注入rhAPC(100ug/kg),D1、D2組在同一時(shí)間點(diǎn)注入同等量的生理鹽水。分離胃粘膜上皮細(xì)胞,并調(diào)制成細(xì)胞濃度為1×106個(gè)/ml的單細(xì)胞懸液,加入DiOC6,上流式細(xì)胞儀檢測(cè)線粒體跨膜電位(Δψm)。采用ELISA法檢測(cè)各組血清中TNF-α、IL-1β、IL-6的濃度。取胃底腺部分胃粘膜上皮組織,10%甲醛固定48小時(shí)后,經(jīng)HE染色,光鏡下觀察。 結(jié)果在WRS各時(shí)間點(diǎn)UI,TNF-α、IL-6的濃度均較正常組明顯升高(P0.01),在WRS2h后TNF-α(647.8±87.55pg/ml)、IL-6(52.85±5.51pg/ml)比WRS3.5h后TNF-α(474.3±103.6)、IL-6(35.6±5.72)為高;胃粘膜的病理?yè)p害以WRS3.5h后更明顯。線粒體跨膜電位較正常組明顯下降,WRS2h后為78.80±33.19,WRS3.5h后為52.83±20.89;IL-1β較正常組有下降趨勢(shì),但無(wú)顯著差異(P0.05)。rhAPC治療組各時(shí)間點(diǎn)TNF-α、IL-6的濃度均較模型組顯著下降(P0.001), IL-1β的濃度較模型組顯著升高(P0.01),線粒體跨膜電位較模型組顯著升高(P0.01),胃粘膜的損傷得到減
[Abstract]:Objective to investigate the protective and therapeutic effects of activated protein (APC) on acute gastric mucosal injury.The model of water-immersion restraint stress (WRS) was established and the ulcer index was calculated. The ulcer index was calculated. Half an hour before the onset of WRS, the rats were injected with the same amount of normal saline through the tail vein of rhAPC-100ugr / kg ~ (-1) D _ (1) D _ (2) group at the same time.Gastric mucosal epithelial cells were isolated and single cell suspensions of 1 脳 106 / ml were modulated. DiOC6 was added. Mitochondrial transmembrane potential (螖 蠄 m) was measured by upflow cytometry.The concentration of TNF- 偽 and IL-1 尾 and IL-6 in serum of each group was detected by ELISA method.The gastric fundus gland was fixed with 10% formaldehyde for 48 hours, then stained with HE and observed under light microscope.Compared with the normal group, the mitochondrial transmembrane potential decreased significantly from 78.80 鹵33.19 WRS3.5h to 52.83 鹵20.89mil IL-1 尾 after 2h compared with the normal group.However, there was no significant difference in the concentration of TNF- 偽 and IL-6, the concentration of IL-1 尾 in the model group was significantly higher than that in the model group, the mitochondrial transmembrane potential was significantly higher than that in the model group, and the gastric mucosal damage was decreased.
