EGF對(duì)乳腺干細(xì)胞體外增殖分化的影響及MSC體內(nèi)移植的實(shí)驗(yàn)研究

【摘要】: 乳腺干細(xì)胞(mammary stem cells,MSCs)是一種存在于乳腺組織中的處于靜止期的,未分化的細(xì)胞,并保持長期自我更新的能力,具有分化為乳腺組織特定形態(tài)特征和獨(dú)特功能的各種類型細(xì)胞。乳腺干細(xì)胞的特點(diǎn)是不對(duì)稱分裂,子代細(xì)胞保持干細(xì)胞特性或開始分化。干細(xì)胞還能對(duì)稱分裂,兩個(gè)子代細(xì)胞均為干細(xì)胞。這提示存在一種機(jī)制調(diào)節(jié)乳腺干細(xì)胞的自我更新能力與分化能力的平衡。乳腺干細(xì)胞可以分化為導(dǎo)管上皮細(xì)胞和肌上皮細(xì)胞。其細(xì)胞專一性的形態(tài)和各專一性的表現(xiàn)及功能特征都是細(xì)胞分化的結(jié)果。目前對(duì)于乳腺干細(xì)胞的研究需要找到能反應(yīng)其細(xì)胞分化及代謝狀況等精細(xì)變化特點(diǎn)的標(biāo)志物,以及分離、純化乳腺干細(xì)胞的技術(shù)手段和調(diào)控增殖與定向分化的各種體外培養(yǎng)體系等。 本研究在第一部分實(shí)驗(yàn)中通過免疫磁珠分離技術(shù)分離出MUC-ESA+的成體乳腺干細(xì)胞,并在體外分離培養(yǎng)及傳代,觀察EGF對(duì)乳腺干細(xì)胞的增殖作用及乳腺干細(xì)胞表面標(biāo)志物的變化。在第二部分實(shí)驗(yàn)中將其應(yīng)用于裸鼠移植實(shí)驗(yàn)觀察其在體內(nèi)的增殖過程,從而為乳腺干細(xì)胞的進(jìn)一步研究和臨床應(yīng)用打下基礎(chǔ)。
[Abstract]:Mammary stem cells (MSCs) is a kind of cells in the mammary gland, which is in the stationary stage, undifferentiated, and maintains the ability of self-renewal for a long time. It has the ability to differentiate into the specific morphological features and unique functions of the mammary gland.Breast stem cells are characterized by asymmetric division, and progeny cells maintain stem cell identity or begin to differentiate.Stem cells also divide symmetrically, and both progeny cells are stem cells.This suggests that there is a mechanism to regulate the balance between self-renewal and differentiation of breast stem cells.Breast stem cells can differentiate into ductal epithelial cells and myoepithelial cells.The morphology of its cell specificity and the expression and functional characteristics of each specificity are the result of cell differentiation.At present, the study of mammary stem cells needs to find markers that can reflect the characteristics of cell differentiation and metabolism, as well as isolation.Methods of purifying mammary stem cells and in vitro culture systems for regulating proliferation and directional differentiation.In the first part of the study, the adult mammary stem cells of MUC-ESA were isolated by immunomagnetic bead separation, and cultured and subcultured in vitro to observe the proliferative effect of EGF on breast stem cells and the changes of surface markers of breast stem cells.In the second part of the experiment, it was applied to the nude mice transplantation experiment to observe its proliferation in vivo, so as to lay a foundation for the further study and clinical application of mammary stem cells.
