【摘要】:背景: 據(jù)調(diào)查發(fā)現(xiàn),,在過去五十年中男性精子質(zhì)量呈下降趨勢,少精癥、無精癥患者屢見不鮮,環(huán)境因素被認(rèn)為是致病的主要因素,其中溫度作為一種外在因素引起男性不育愈來愈被重視,研究表明:包括人類在內(nèi)的大部分哺乳動(dòng)物的睪丸需要一個(gè)比腹腔溫度低的環(huán)境,睪丸暴露在腹腔溫度或高于腹腔溫度可導(dǎo)致一種可復(fù)性的生精障礙和生精細(xì)胞退化,可引起暫時(shí)的、部分或完全的不育。這種高溫作用在不同的生長發(fā)育階段,對(duì)其產(chǎn)生的影響可能是不同的。青春期是男性發(fā)育過程中第二個(gè)高峰期,在此期男性會(huì)出現(xiàn)第二性征并且生殖器官漸趨成熟,青春期對(duì)男性不僅在身體發(fā)育方面還是在性腺發(fā)育方面都具有重要意義,如此期受到外界環(huán)境不良干擾后可能會(huì)影響男性生殖功能和個(gè)體的生長發(fā)育。現(xiàn)今,男性不育患者日益增多,往往都是成年后才會(huì)發(fā)現(xiàn),從而尋找原因給予相應(yīng)的治療,但一部分不育患者可能是在嬰幼年或青少年期受到某種不良刺激后而引起的。目前隨著醫(yī)療水平的提高,男性醫(yī)學(xué)保健教育的廣泛宣傳,男性對(duì)自己的生殖健康提高重視,出現(xiàn)男性不育癥愈來愈年輕化的現(xiàn)象。另外,越來越多的研究報(bào)道男性不育和氧化損傷有著密切的關(guān)系,活性氧是導(dǎo)致男性不育的罪魁禍?zhǔn)。因此,本研究試圖探討青春期前后大鼠睪丸局部高溫對(duì)青春期前和青春期生精細(xì)胞凋亡的影響,同時(shí)并探討生精細(xì)胞發(fā)生凋亡、氧化應(yīng)激、睪酮三者之間的關(guān)系。 目的: 1) 研究青春期前后大鼠睪丸局部熱作用后睪丸生精上皮的變化; 2) 探索睪丸局部熱作用后睪丸組織氧化應(yīng)激作用及其與生精細(xì)胞凋亡的關(guān)系; 3) 研究睪丸局部熱作用后睪酮的變化及其與氧化應(yīng)激的關(guān)系。 材料與方法: 雄性Sprague-Dawley(SD)青春期前大鼠(5周齡),分籠飼養(yǎng)于標(biāo)準(zhǔn)動(dòng)物房(22℃),給予規(guī)律光照(12h∶12h),自由飲食,隨機(jī)分熱處理組和對(duì)照組,熱處理組按熱處理后2h、
[Abstract]:Background:According to the survey, in the past 50 years, the quality of male sperm has been declining, oligozoospermia and azoospermia are common, and environmental factors are considered to be the main cause of the disease.Temperature, as an external factor, has attracted increasing attention to male infertility. Studies have shown that the testicles of most mammals, including humans, need an environment with lower temperatures than that of the abdominal cavity.Testicular exposure to or above celiac temperature may lead to a refolding of spermatogenic disorders and degeneration of spermatogenic cells, which may cause temporary, partial or complete infertility.The effect of this high temperature on it may be different at different stages of growth and development.Adolescence is the second peak of male development, in which men develop secondary sexual symptoms and mature reproductive organs. Adolescence is of great significance to men not only in physical development but also in gonadal development.This period may affect male reproductive function and individual growth and development after being disturbed by external environment.Nowadays, the increasing number of male infertility patients are often found in adulthood, so as to find a reason to give corresponding treatment, but some infertility patients may be caused by some adverse stimulation in infancy and adolescence.At present, with the improvement of medical level and the widespread propaganda of male medical health education, men pay more attention to their reproductive health, and the phenomenon of male infertility becoming younger and younger.In addition, more and more studies have reported that male infertility and oxidative damage are closely related, reactive oxygen species (Ros) is the main cause of male infertility.Therefore, this study attempts to explore the effects of local high temperature of testis before and after puberty on apoptosis of spermatogenic cells before and after puberty, and to explore the relationship among apoptosis of spermatogenic cells, oxidative stress and testosterone.Objective:1) to study the changes of testicular spermatogenic epithelium after local heat treatment in rats before and after puberty.2) to explore the relationship between oxidative stress and spermatogenic cell apoptosis in testicular tissue after local heat treatment.3) to study the changes of testosterone and the relationship between testosterone and oxidative stress.Materials and methods:Male Sprague-Dawley (SD) prepubertal rats of 5 weeks of age were raised in cage at 22 鈩
