骨髓多能成體祖細(xì)胞向表皮細(xì)胞分化的研究

【摘要】: 本研究通過克隆化培養(yǎng),成功的從成年大鼠骨髓中分選出較為均一的具有多系分化潛能的多能成體祖細(xì)胞(MAPCs)群體。首次系統(tǒng)比較了骨髓中間充質(zhì)干細(xì)胞(MSCs)和MAPCs在生物特性及分化潛能上的異同點(diǎn)。結(jié)果證實(shí),MAPCs可能是MSCs更早期的一類遠(yuǎn)祖細(xì)胞;體外模擬MAPCs向皮膚上皮細(xì)胞分化的微環(huán)境,首次建立了MAPCs向表皮細(xì)胞體外分化模型,實(shí)現(xiàn)MAPCs向表皮樣細(xì)胞的轉(zhuǎn)型;在體觀察了皮膚局部損傷微環(huán)境對MAPCs的遷移和誘導(dǎo)分化作用。結(jié)果提示MAPCs可定向遷移受損皮膚表面及皮膚附屬器毛囊周圍,并通過轉(zhuǎn)化為表皮細(xì)胞參與受損皮膚的愈合。該工作不僅豐富了MAPCs可塑性基礎(chǔ)理論研究,而且為指導(dǎo)細(xì)胞移植治療應(yīng)用到臨床實(shí)踐提供了一定的理論依據(jù)。
[Abstract]:In this study, a more homogeneous multifunctional adult progenitor cell (MAPCs) with multilineage differentiation potential was successfully isolated from adult rat bone marrow by cloning culture.Bone marrow mesenchymal stem cells (MSCs) and MAPCs were compared in biological characteristics and differentiation potential for the first time.The results showed that MPCs might be an earlier type of MSCs progenitor cells, and mimic the microenvironment of MAPCs differentiation into skin epithelial cells in vitro. The model of MAPCs differentiation into epidermal cells was established for the first time to realize the transition of MAPCs to epidermoid cells.The effects of local skin damage on MAPCs migration and differentiation were observed in vivo.The results suggest that MAPCs can migrate the damaged skin surface and the skin adnexa around the hair follicle and participate in the healing of the damaged skin by transforming into epidermal cells.This work not only enriches the basic theory of MAPCs plasticity, but also provides a theoretical basis for the application of cell transplantation therapy to clinical practice.
