【摘要】：restin基因是1999年本室在研究全反式維甲酸誘導(dǎo)HL-60細(xì)胞分化時(shí)克隆得到的一個(gè)人類未知功能的新基因,因其轉(zhuǎn)錄物在維甲酸誘導(dǎo)凋亡的細(xì)胞中表達(dá)上調(diào),因此最初被命名為凋亡相關(guān)基因Apr-1(apoptosis related gene);2001年,Chomez等通過同源序列篩選Genbank數(shù)據(jù)庫(kù),也發(fā)現(xiàn)了該基因,并命名為melanoma associated antigen H1(MAGE-H1)。 restin基因編碼蛋白由219個(gè)氨基酸組成,為堿性核蛋白;N端含一段細(xì)胞核定位信號(hào),在氨基酸序列的中央偏C端含一個(gè)MAGE家族的保守結(jié)構(gòu)域(MAGE homology domain,MHD);restin基因定位于Xp11.22,開放讀框全部位于一個(gè)外顯子中;從基因進(jìn)化樹分析發(fā)現(xiàn),restin與Ⅱ類MAGE家族成員比較靠近;組織表達(dá)譜分析表明Restin主要表達(dá)在正常終末分化組織,在腦、睪丸和胎兒組織中表達(dá)最高,在外周血細(xì)胞中表達(dá)較低,而在腫瘤組織中不表達(dá)或表達(dá)量極低。
[Abstract]:Restin gene is a novel gene cloned in 1999 to study the differentiation of HL-60 cells induced by all-trans retinoic acid (ATRA).Therefore, it was originally named Apr-1(apoptosis related gene, which was identified as melanoma associated antigen H1 MAGE-H1 by homologous sequence screening of Genbank database in 2001.The protein encoded by restin gene consists of 219 amino acids.In the C-terminal of the amino acid sequence, a conserved domain of the MAGE family, mage homology domain, MHDHDStin gene was located in Xp11.22, and the open reading frame was all located in one exon.Tissue expression profile analysis showed that Restin was mainly expressed in normal terminal differentiated tissues, the highest in brain, testis and fetal tissues, but low in peripheral blood cells, but not in tumor tissues.
