人牙髓干細胞的體外培養(yǎng)及神經(jīng)樣誘導分化

【摘要】:背景:牙髓組織來源干細胞的發(fā)現(xiàn)及概念的確立有助于從細胞水平上認識牙齒發(fā)育和再生修復機制。目的:了解人牙髓干細胞向神經(jīng)元樣細胞誘導分化能力和誘導分化條件。方法:取健康青年人的第三磨牙的牙髓組織后,制成單細胞懸液,加入含有體積分數(shù)為15%胎牛血清α-MEM培養(yǎng)基在6孔板培養(yǎng),傳代培養(yǎng)后加入丁羥基茴香醚、forskolin、β-巰基乙醇、堿性成纖維細胞生長因子誘導劑進行培養(yǎng)。結果與結論:免疫熒光和反轉錄-聚合酶鏈反應檢測顯示,誘導2周后人牙髓細胞除了stro-1,Col-Ⅰ,牙本質(zhì)涎蛋白表達陽性外,還有巢蛋白,神經(jīng)元特異性烯醇化酶的表達也是陽性,而牙齦纖維細胞表達均為陰性。說明人牙髓組織中存在成體干細胞,在一定的誘導培養(yǎng)條件下,牙髓干細胞有向神經(jīng)元樣細胞分化的潛能。
[Abstract]:Background: the discovery of dental pulp tissue derived stem cells and the establishment of their concepts are helpful to understand the mechanism of tooth development and regeneration at the cellular level. Objective: to understand the ability and inducement of human dental pulp stem cells to differentiate into neuron-like cells. Methods: the pulp tissue of the third molar of healthy young people was taken. Single cell suspension was prepared, and 偽 -MEM medium containing 15% fetal bovine serum was added in 6-well plate culture medium. After subculture, butyl hydroxyl anisole forskolin, 尾 -mercaptoethanol were added.
