【摘要】：目的：探討雷公藤多甙造成雌性育齡期小鼠生殖功能低下的藥理作用機(jī)理，建立雷公藤多甙致雌性小鼠生殖功能低下動(dòng)物模型。 方法：采用7～8周齡雌性成熟昆明種小鼠，通過(guò)陰道細(xì)胞學(xué)涂片篩選出有正常動(dòng)情周期者，隨機(jī)分為雷公藤多甙A、B、C、D、E5個(gè)劑量組及正常對(duì)照組，分別給予雷公藤多甙60mg／kg／d、40mg／kg／d、30mg／kg／d、20mg／kg／d、15mg／kg／d及生理鹽水。連續(xù)每日觀察給藥后小鼠動(dòng)情周期狀態(tài)，根據(jù)動(dòng)情周期變化情況推斷給藥后小鼠的生殖功能狀態(tài)。分別在①各組部分小鼠開始出現(xiàn)動(dòng)情周期延長(zhǎng)，②各組小鼠動(dòng)情周期均延長(zhǎng)、不規(guī)則，③各組小鼠動(dòng)情周期完全紊亂、甚至消失三個(gè)時(shí)段分批處死部分小鼠，以觀察雷公藤多甙在不同時(shí)段對(duì)小鼠生殖功能的影響。采用放射免疫法檢測(cè)血清E_2、P水平，組織稱重法計(jì)算卵巢、子宮指數(shù)，病理切片觀察各時(shí)段卵巢、子宮組織形態(tài)學(xué)變化，計(jì)數(shù)各級(jí)卵泡、黃體數(shù)，測(cè)量子宮內(nèi)膜、肌層厚度。 結(jié)果：給藥5周時(shí)，各組部分小鼠出現(xiàn)動(dòng)情周期延長(zhǎng)；給藥7周時(shí)，各組小鼠均出現(xiàn)動(dòng)情周期延長(zhǎng)、不規(guī)則；給藥10周時(shí)，各組小鼠動(dòng)情周期全部紊亂，部分動(dòng)情周期消失，分別在5、7、10周時(shí)分批處死小鼠。結(jié)果顯示：給藥5周、7周時(shí)，雷公藤多甙不能降低E_2、P水平、卵巢指數(shù)和卵泡、黃體數(shù)，給藥7周時(shí)A組子宮指數(shù)降低，內(nèi)膜、肌層變薄。給藥10周時(shí)，，A、B、C組子宮指數(shù)明顯降低，子宮內(nèi)膜、肌層明顯變薄，卵巢成熟卵泡數(shù)及黃體數(shù)減少(P＜0.05或P＜0.01)；A、B組E_2水平降低(P＜0.05)，各組P水平和卵巢指數(shù)無(wú)明顯變化。 結(jié)論：1．雷公藤多甙給藥10周時(shí)60mg／kg／d、40mg／kg／d及30mg／kg／d劑量均能在一定程度上損傷小鼠生殖功能，造成腎虛生殖功能低下狀態(tài)。2．建議用雷公藤多甙30mg／kg／d給藥10周以建立雌性小鼠腎虛生殖功能低下模型。
[Abstract]:Objective: to investigate the pharmacological mechanism of female reproductive dysfunction induced by tripterygium wilfordii polyglycosides, and to establish an animal model of female reproductive dysfunction induced by tripterygium wilfordii polyglycosides. Methods: female adult Kunming mice aged 7 to 8 weeks were selected by vaginal cytological smears. Those with normal estrous cycle were randomly divided into 5 dose group and normal control group of Tripterygium wilfordii. They were given 60 mg / kg / kg of Tripterygium wilfordii, 40 mg / kg / kg / d, 30 mg / kg / kg / d, 20 mg / kg / kg / d and 15 mg / kg / d of normal saline, respectively. The oestrous cycle of mice was observed continuously after administration of the drug daily. According to the changes of estrus cycle, the reproductive function of mice after administration was inferred. In order to observe the effect of tripterygium wilfordii polyglycosides on the reproductive function of mice, the serum E2P level was detected by radioimmunoassay, and the ovarian and uterine index was calculated by tissue weighing method. Histologic changes of ovary and uterus were observed by pathological section. Follicles and luteal bodies were counted and the thickness of endometrium and myometrium were measured. Results: at 5 weeks of administration, the estrus cycle of some mice in each group was prolonged; at 7 weeks, the estrus cycle was prolonged and irregular in all the mice of each group; at 10 weeks of administration, the estrus cycle of all the mice in each group was disordered, and part of the estrus cycle disappeared. Mice were killed in batches at 10 weeks after 5 weeks of administration. The results showed that tripterygium wilfordii polyglycosides could not decrease E2P level, ovarian index, follicle and corpus luteum at 5 weeks and 7 weeks after administration, and the uterine index and endometrium of group A were decreased at 7 weeks after administration of tripterygium wilfordii. After 10 weeks of administration, the uterine index, endometrium, myometrium and the number of mature follicles and luteal bodies decreased significantly (P < 0.05 or P < 0.01). Conclusion: 1. The dose of 60 mg / kg / kg / d and 30 mg / kg / d of tripterygium wilfordii can damage the reproductive function of mice to a certain extent after 10 weeks of administration of tripterygium wilfordii. It was suggested that 30 mg / kg / d of Tripterygium wilfordii polyglycosides should be given for 10 weeks in order to establish the model of low reproductive function in female mice with kidney deficiency.
