皮質(zhì)顆粒在評(píng)估體外受精中卵母細(xì)胞胞質(zhì)成熟度作用的研究

【摘要】:近年來(lái)一些研究認(rèn)為卵母細(xì)胞胞核和胞質(zhì)成熟程度,尤其是胞質(zhì)成熟度是影響體外受精—胚胎移植(in vitro fertilization-embryo transfer,IVF-ET)成功率的一個(gè)重要原因。卵母細(xì)胞成熟度包括胞核成熟和胞質(zhì)成熟,第一極體(first body,PB1)是明顯的可以直接觀察到的細(xì)胞核成熟的特征,而如何觀察和評(píng)價(jià)卵母細(xì)胞胞質(zhì)成熟程度,進(jìn)而分析受精失敗的原因,一直是我們面臨的難題;目前大多數(shù)學(xué)者把皮質(zhì)顆粒(cortical granule,CG)的分布和遷移視為評(píng)價(jià)胞質(zhì)成熟度的一項(xiàng)重要指標(biāo)。CG的皮質(zhì)反應(yīng)和透明帶反應(yīng)是保證正常受精和阻止多精受精的兩個(gè)重要生理功能;CG的這些特性,既是細(xì)胞質(zhì)成熟的一個(gè)標(biāo)志,也是細(xì)胞質(zhì)成熟的重要部分;所以本研究將CG在卵母細(xì)胞質(zhì)膜下的線性排列作為細(xì)胞質(zhì)成熟的指標(biāo)。關(guān)于人卵母細(xì)胞CG的分布情況,國(guó)內(nèi)尚未見到報(bào)道。本研究以CG作為評(píng)價(jià)胞質(zhì)成熟的指標(biāo),一方面以昆明小白鼠為模型,研究了卵母細(xì)胞成熟過(guò)程中CG的遷移和排放情況,證實(shí)了CG的遷移及分布情況與卵母細(xì)胞的成熟度有著直接的關(guān)系,在保證正常受精及阻止多精受精方面發(fā)揮了重要作用;另一方面通過(guò)熒光顯微鏡觀察了人IVF周期中未成熟、未受精、多精受精卵及發(fā)育阻滯的合子中CG的分布和遷移情況,對(duì)卵母細(xì)胞進(jìn)行了胞質(zhì)成熟度的評(píng)估。本研究旨在快速判斷卵母細(xì)胞的成熟度,找出常規(guī)IVF第一周期受精率低或受精失敗的原因,為患者以后的治療提供基礎(chǔ)與臨床上的理論依據(jù),從而協(xié)助患者選擇下一周期的最佳治療方法、受精方式和體外培養(yǎng)體系。
[Abstract]:In recent years, some studies have suggested that oocyte nuclear and cytoplasmic maturation, especially cytoplasmic maturity, is an important factor affecting the success rate of in vitro fertilization and embryo transfer in vitro fertilization-embryo transfer IVF-ET.The maturation of oocytes includes nuclear maturation and cytoplasmic maturation. The first polar body first body (PB1) is an obvious characteristic of nuclear maturation which can be observed directly. However, how to observe and evaluate the cytoplasmic maturation of oocytes and then analyze the causes of fertilization failure has been a difficult problem. At present, most scholars regard the distribution and migration of cortical granuleus (CGG) as an important index for evaluating cytoplasmic maturity. CG's cortical reaction and pellucida response are two important physiological tasks to ensure normal fertilization and prevent multiple fertilization. These are the properties of CG, It is not only a marker of cytoplasmic maturation, but also an important part of cytoplasmic maturation. In this study, the linear arrangement of CG under oocyte plasma membrane was taken as the index of cytoplasmic maturation. In this study, CG was used as an index to evaluate cytoplasmic maturation. On the one hand, the migration and emission of CG during oocyte maturation were studied using Kunming mice as a model. It was proved that the migration and distribution of CG was directly related to the maturation of oocytes, which played an important role in ensuring normal fertilization and preventing poly-spermatid fertilization, on the other hand, the immature of human IVF cycle was observed by fluorescence microscope. The distribution and migration of CG in unfertilized, polysperm fertilized eggs and developmental retarded zygotes were evaluated to evaluate the cytoplasmic maturity of oocytes. To find out the causes of low fertilization rate or failure of fertilization in the first cycle of conventional IVF, to provide basic and clinical theoretical basis for the subsequent treatment of patients, so as to assist patients to select the best treatment method, fertilization method and in vitro culture system for the next cycle.
