冠脈結(jié)扎法建立大鼠急性心肌梗死模型方法的探討

【摘要】:目的建立一種方便、省時(shí)、死亡率低且穩(wěn)定的急性心肌梗死模型制作方法。方法雄性成年Sprague·Dawley大鼠麻醉后在呼吸機(jī)支持下,于相應(yīng)第3、4脅間隙處開胸,擠壓兩側(cè)胸壁,將心臟擠出于胸腔外,通過(guò)結(jié)扎左冠狀動(dòng)脈前降支,建立心肌梗死模型。通過(guò)觀察術(shù)后大鼠心電圖的變化,以及分別利用Evan blue染色及病理組織學(xué)方法于術(shù)后3 h及1周,確定心肌缺血及梗死范圍,同時(shí)觀察術(shù)中、術(shù)后24 h及1周大鼠的存活率。結(jié)果術(shù)后大鼠Ⅰ、Ⅱ?qū)?lián)心電圖ST段明顯抬高。術(shù)后3 h大鼠心臟Evan blue染色顯示心肌缺血區(qū)恒定。術(shù)后1周處死取出大鼠心臟,可見心臟前壁、側(cè)壁及心尖處形成境界清晰的心肌梗死區(qū),梗死區(qū)顏色明顯淺于周圍的心肌,呈蒼白色,失去正常光澤。術(shù)后1周大鼠心肌組織HE染色,可見梗死區(qū)伴有大量炎性細(xì)胞浸潤(rùn)。造模成功率為83%,術(shù)后24 h存活率為77%,術(shù)后1周大鼠總存活率為72%。結(jié)論此模型操作簡(jiǎn)單、穩(wěn)定,且術(shù)后動(dòng)物存活率高,是研究心肌梗死較理想的模型。
[Abstract]:Objective to establish a convenient, time-saving, low mortality and stable model of acute myocardial infarction (AMI). Methods male adult Sprague 路Dawley rats were anesthetized and treated with ventilator. The myocardial infarction model was established by ligating the anterior descending branch of the left coronary artery by squeezing the heart out of the chest. The changes of electrocardiogram (ECG) of the rats were observed after operation, and Evan blue staining and histopathology were used at 3 h and 1 week after operation, respectively.
