【摘要】：免疫球蛋白(IgG)是免疫系統(tǒng)中的重要分子，以往經(jīng)典的免疫學理論認為IgG都是B淋巴細胞和漿細胞分泌的，而新近的研究表明上皮性腫瘤也能分泌IgG。同時以往的研究已證實，在腫瘤細胞中，熱休克蛋白(HSP)高表達。我們在實際工作中發(fā)現(xiàn)IgG和HSP70的免疫組化染色表現(xiàn)非常相似，于是我們設(shè)想IgG和HSP70也許有相互關(guān)系，并在腫瘤免疫中互相作用。 本課題旨在探討：腫瘤細胞分泌的IgG與熱休克蛋白70(HSP70)的共定位關(guān)系，并初步提出在腫瘤細胞內(nèi)可能存在細胞內(nèi)免疫機制，，討論IgG和HSP70在細胞內(nèi)免疫系統(tǒng)中的作用。目的：1．制備IgG的多克隆抗體；2．制備IgG的單克隆抗體；3．研究證實IgG和HSP70在腫瘤細胞中的共定位關(guān)系，為腫瘤細胞內(nèi)免疫機制的存在提供初步證據(jù)。方法：1．用純化的IgG以常規(guī)多克隆抗體制備方法制備兔抗人IgG的多克隆抗體，并純化并對抗體特性進行鑒定；2．以純化的IgG常規(guī)免疫、常
[Abstract]:Immunoglobulin G (IgG) is an important molecule in the immune system. The classical immunological theory holds that IgG is secreted by B lymphocytes and plasma cells, but recent studies have shown that epithelial tumors can also secrete IgG.At the same time, previous studies have confirmed that IgG can be secreted by B lymphocytes and plasma cells. In tumor cells, heat shock protein (HSP) is highly expressed. We found that IgG and HSP70 showed very similar immunohistochemical staining, so we assumed that IgG and HSP70 might be related to each other and interact with each other in tumor immunity. The aim of this study was to investigate the co-localization of IgG secreted by tumor cells and heat shock protein 70 (HSP70), and to suggest that there may be intracellular immune mechanisms in tumor cells. To discuss the role of IgG and HSP70 in the cellular immune system. Objective 1. To prepare the polyclonal antibody to IgG. 2. To prepare the monoclonal antibody against IgG. The co-localization of IgG and HSP70 in tumor cells was confirmed.
