血管內(nèi)皮抑制因子angiostatin和thalidomide對(duì)淋巴管內(nèi)皮細(xì)胞生成的影響

【摘要】:目的 探討血管生成抑制素(angiostatin)和反應(yīng)停(thalidomide)對(duì)淋巴管內(nèi)皮細(xì)胞增殖和游走的影響及作用機(jī)制。旨在找到安全、有效、實(shí)用的淋巴管生成抑制劑。 材料和方法 實(shí)驗(yàn)所用的淋巴管內(nèi)皮細(xì)胞取自豬的胸導(dǎo)管。1、抑制實(shí)驗(yàn):本實(shí)驗(yàn)分為兩大組:angiostatin組和thalidomide組。每大組分別采用劃線法和MTT法觀察兩種因子對(duì)淋巴管內(nèi)皮細(xì)胞增殖和游走的影響。(1)劃線法:angiostatin和thalidomide組分別包括各自的對(duì)照組和3種濃度的實(shí)驗(yàn)組。(2)MTT法:angiostatin組包括對(duì)照組和4種濃度的實(shí)驗(yàn)組;thalidomide組包括對(duì)照組和5種濃度的實(shí)驗(yàn)組。2、細(xì)胞凋亡實(shí)驗(yàn):(1)Hoechst染色法。(2)流式細(xì)胞儀檢測(cè)法:配置不同濃度的angiostatin和thalidomide作用于淋巴管內(nèi)皮細(xì)胞檢測(cè)細(xì)胞的凋亡率。3電鏡觀察。 結(jié)果 1、劃線法:與對(duì)照組相比,當(dāng)angiostatin和thalidomide的濃度達(dá)到0.5μg/ml時(shí),兩者對(duì)淋巴管內(nèi)皮細(xì)胞的增殖和游走都有明顯的抑制作用(P0.01)。2、MTT法:當(dāng)angiostatin和thalidomide的濃度分別達(dá)到0.75μg/ml和0.5μg/ml時(shí),兩者對(duì)內(nèi)皮細(xì)胞的增殖具有顯著的抑制作用(P0.01)。3、凋亡實(shí)驗(yàn):(1)Hoechst染色證實(shí),經(jīng)angiostatin和thalidomide處理后的淋巴管內(nèi)皮細(xì)胞,可觀察到其核周圍有凋亡小體。(2)流式細(xì)胞儀檢測(cè)顯示出大量亞二倍體細(xì)胞核型峰(凋亡峰),二倍體細(xì)胞峰減少。 結(jié)論 1、angiostatin和thalidomide對(duì)淋巴管內(nèi)皮細(xì)胞的增殖和游走具有明顯的抑制作用,且有劑量依賴性。2、angiostatin和thalidomide具有促進(jìn)細(xì)胞凋亡的作用。
[Abstract]:Objective to study angiostatin (angiostatin) and thalidomide in response to angiogenesis. The effect and mechanism on proliferation and migration of lymphatic endothelial cells. Effective, practical lymphangiogenesis inhibitor. Materials and methods the lymphatic endothelial cells used in the experiment were taken from the thoracic ducts of pigs. Inhibition experiment: this experiment is divided into two groups:. In angiostatin group and thalidomide group, the effects of two factors on the proliferation and migration of lymphatic endothelial cells were observed by underlined method and MTT method respectively. 1) the two groups: the control group and the thalidomide group included their respective control groups and three different concentrations of experimental groups (n = 2). MTT method: angiostatin group included control group and four experimental groups of different concentrations. The thalidomide group included control group and experimental group of 5 different concentrations. Apoptosis experiment: 1: 1 Hoechst staining. Flow cytometry: the apoptotic rate of lymphatic endothelial cells was observed by electron microscope with different concentrations of angiostatin and thalidomide. Results 1. Line method: compared with the control group, when the concentration of angiostatin and thalidomide reached 0.5 渭 g / ml. Both of them had obvious inhibitory effects on proliferation and migration of lymphatic endothelial cells. MTT method: when the concentration of angiostatin and thalidomide reached 0.75 渭 g / ml and 0.5 渭 g / ml, respectively. Both of them had a significant inhibitory effect on endothelial cell proliferation. The apoptosis experiment was confirmed by Hoechst staining. The lymphatic endothelial cells were treated with angiostatin and thalidomide. It was observed that there were apoptotic corpuscles around its nucleus. Flow cytometry showed that there were a large number of nuclear peaks (apoptosis peak and diploid cell peak decreased) in hypodiploid cells. Conclusion 1 angiostatin and thalidomide can inhibit the proliferation and migration of lymphatic endothelial cells in a dose-dependent manner. Angiostatin and thalidomide can promote apoptosis.
