人芽囊原蟲形態(tài)學(xué)和致病性的研究

【摘要】:背景和目的 人芽囊原蟲(Blastocystis hominis, B. hominis)是一種越來越引起學(xué)者們關(guān)注的腸道寄生性原蟲。自1911由Alexief首次發(fā)現(xiàn)以來,由于B. hominis具有形態(tài)多變及光鏡下不宜辨認(rèn)等特點,一直未引起學(xué)術(shù)界的普遍重視。我國在B. hominis研究方面起步較晚,1990年以來雖然少數(shù)學(xué)者曾進(jìn)行了形態(tài)學(xué)、病理學(xué)、區(qū)域流行病學(xué)方面的研究,但各研究仍屬起步階段。對B. hominis進(jìn)一步實驗研究很有必要。 本研究結(jié)合B. hominis獨特的生化待點和可能的生活周期,用多種普通和特殊的染色方法對B. hominis的形態(tài)進(jìn)行了光鏡、電鏡的觀察,以進(jìn)一步了解其形態(tài)特點,并結(jié)合形態(tài),對B. hominis某些細(xì)胞器功能進(jìn)行了初步探討。然后將培養(yǎng)的不同數(shù)量的空泡型B. hominis感染免疫功能正常和免疫功能低下小鼠,比較不同數(shù)量感染的差異,,免疫功能正常及免疫功能低下小鼠感染B. hominis的差異。小鼠感染實驗及建立B. hominis的動物感染模型是為研究B. hominis的致病性及致病機理奠定了基礎(chǔ)。 材料和方法 1.形態(tài)學(xué)觀察 本實驗用B. hominis陽性的糞便標(biāo)本接種于加有10%滅活胎牛血清的locke氏-雞蛋雙相培養(yǎng)基(LES)和1640培養(yǎng)基(RPMI-1640)培養(yǎng),37℃分離培養(yǎng),48-72h轉(zhuǎn)種一次傳代保種,轉(zhuǎn)種時加入青霉素和鏈霉素。轉(zhuǎn)種后每日吸取培養(yǎng)液經(jīng)碘液染色和H.E染色,光鏡下觀察蟲體各期形態(tài)并拍照。同時將培養(yǎng)液制備成電鏡標(biāo)本,置掃描電鏡下觀察各期蟲體的超微形態(tài)結(jié)構(gòu)。 2.B. hominis感染小鼠
[Abstract]:Background and purpose Blastocystis hominis. B. hominis is an intestinal parasitic protozoa that has attracted increasing attention from researchers since it was first discovered by Alexief in 1911. Because of its characteristics of changeable morphology and unsuitable recognition under light microscope, B. hominis has not been paid much attention to in academic circles. Our country started late in the research of B. hominis. Since 1990, a few scholars have studied morphology, pathology, and regional epidemiology. However, the studies are still at the initial stage. It is necessary to further study the B. Hominis experiment. In this study, the morphology of B. hominis was examined by various ordinary and special staining methods combined with the unique biochemistry of B. hominis and its possible life cycle. Electron microscope observation to further understand its morphological characteristics, and combined with morphology. The function of some organelles of B. hominis was preliminarily studied. Then the vacuolar type B. Hominis was infected with normal immune function and hypoimmune function in mice. To compare the difference of infection quantity.
