對(duì)實(shí)驗(yàn)性腎臟病動(dòng)物模型的再認(rèn)識(shí)

【摘要】:腎臟病動(dòng)物模型一般分為二類(lèi),一類(lèi)是通過(guò)施加人為操作而模擬人類(lèi)腎臟病的模型,這類(lèi)經(jīng)典的動(dòng)物模型包括抗腎小球基底膜抗體腎炎、Heymann腎炎、抗Thy1.1抗體腎炎、BSA腎炎以及嘌呤霉素腎病等;另一類(lèi)動(dòng)物模型是"自然發(fā)病模型",也就是讓動(dòng)物自然出現(xiàn)近似人類(lèi)腎臟病的病理特征,它們包括HIGA小鼠Ig A腎病和NZB/WF1小鼠、MRL/1pr小鼠狼瘡性腎炎模型。另外,借助基因敲除、siRNA轉(zhuǎn)染等現(xiàn)代分子生物學(xué)技術(shù)也可以制作出轉(zhuǎn)基因動(dòng)物模型。對(duì)于腎臟病領(lǐng)域的中藥藥效學(xué)、藥理學(xué)研究而言,首要的是,正確認(rèn)識(shí)各種腎臟病動(dòng)物模型的特點(diǎn),針對(duì)實(shí)驗(yàn)的具體目的而選擇合適的模型;其次的是,需要病證結(jié)合動(dòng)物模型的支撐。因此,建立具有中醫(yī)證候特征的腎臟病動(dòng)物模型是今后的發(fā)展方向。

Anti Thy1.1 antibody nephritis and purine mycin nephropathy; Another type of animal model is the "natural pathogenesis model", which allows animals to naturally exhibit pathological features similar to human kidney disease, including HIGA mice IgA nephropathy and NZB/WF1 mice. MRL/1pr mice model of lupus nephritis. In addition, gene knockout. Modern molecular biological techniques such as siRNA transfection can also produce transgenic animal models. For the pharmacodynamics and pharmacology of traditional Chinese medicine in the field of kidney disease, the most important thing is. Correct understanding of the characteristics of various animal models of kidney disease, according to the specific purpose of the experiment to select the appropriate model; Secondly, it needs the support of animal model of combination of disease and syndrome. Therefore, the establishment of kidney disease animal model with TCM syndrome characteristics is the development direction in the future.
