SD大鼠數(shù)字解剖數(shù)據(jù)集研究

【摘要】: 如今實驗動物已經(jīng)成為遺傳學、疾病以及藥物開發(fā)等研究領域最基本的研究工具之一。為了方便各種生物學信息的共享和整合,同時出于對動物生存權利及動物福利的考慮,小動物的結構與功能信息的數(shù)字化已經(jīng)成為國內外各研究機構的研究熱點。數(shù)字解剖數(shù)據(jù)集以其更加準確的、直觀的、圖形化表示方法等具有廣泛應用價值的特點,引起了越來越多的科研人員的關注。 本文通過對不同斷層解剖數(shù)據(jù)獲取方式分析、對比,基于冰凍斷層技術搭建了適于小動物研究的斷層解剖成像系統(tǒng)。以SD大鼠為例,構建了SD大鼠斷層解剖數(shù)據(jù)集,對斷層解剖數(shù)據(jù)集的構建方法、工藝路線、技術參數(shù)選擇、可能出現(xiàn)的問題及其解決方法進行了系統(tǒng)的研究。并且基于SD大鼠斷層解剖數(shù)據(jù)集,構建了SD大鼠數(shù)字解剖數(shù)據(jù)集,并對其技術方案和構建技術進行了深入的研究。課題主要實驗結果和結論如下所示: (1)基于冰凍銑削技術搭建了適于小動物研究的斷層解剖成像系統(tǒng),技術參數(shù)為:斷層切削設備的最小進給量為0.001 mm,重復定位精度為0.01/300 mm。該系統(tǒng)具有如下特點,大體積樣品成像;空間分辨率高;侵入型成像方法;斷層圖像具有生物樣品原有的組織紋理、顏色等細節(jié)信息。該系統(tǒng)可以獲得小動物的精確結構信息,為小動物研究提供了一種新的、有效的手段。 (2)本文以兩只成年SD大鼠為例,敘述了斷層解剖成像的實驗過程。并且基于適于小動物研究的斷層解剖成像系統(tǒng),獲取了SD大鼠斷層解剖數(shù)據(jù)集(體素尺寸為20μm×20μm×20μm,圖像分辨率為4,600×2,580×24 bit,BMP格式,共9475張,314.68 GB)和血管灌注的SD大鼠斷層解剖數(shù)據(jù)集(體素尺寸為20μm×20μm×100μm,圖像分辨率為4,917×3,446×24 bit,BMP格式,共1646張,77.9 GB)。 (3)探討了構建數(shù)字解剖數(shù)據(jù)集的技術方案和構建技術,并且基于SD大鼠斷層解剖數(shù)據(jù)集和血管灌注的SD大鼠斷層解剖數(shù)據(jù)集,構建了SD大鼠數(shù)字解剖數(shù)據(jù)集,獲得了高精度的SD大鼠三維解剖結構模型及動脈系統(tǒng)結構模型,具有廣泛的教育及應用價值。 (4)“Visual Rat Atlas”和“Visual Rat Explorer”分別顯示SD大鼠的不同截面的系列斷層圖像、解剖圖譜和SD大鼠的三維解剖結構模型。軟件的使用使得研究人員能夠方便的對數(shù)字解剖數(shù)據(jù)集進行分析和計算,是一種方便實用的研究工具。 研究證明適于小動物研究的斷層解剖成像系統(tǒng),為小動物結構信息研究提供了一種有效的手段;谠撓到y(tǒng)構建的數(shù)字解剖數(shù)據(jù)集可以作為一個框架整合更多生物學信息,研究不同層次、不同水平的生物學信息,為小動物研究及其在生物醫(yī)學研究中提供了應用模型。
[Abstract]:Nowadays , experimental animals have become one of the most basic research tools in genetics , disease and drug development . In order to facilitate the sharing and integration of various biological information , the digitization of the structure and function information of small animals has become a hot topic at home and abroad in order to facilitate the sharing and integration of animal survival rights and animal welfare . In this paper , a tomographic imaging system suitable for small animals has been established by means of analysis and comparison of different anatomical data . Based on SD rats , an anatomical data set of SD rats was constructed . The data set of SD rats was constructed on the basis of SD rats , and its technical scheme and construction techniques were studied . ( 1 ) Based on the frozen milling technology , a tomographic imaging system suitable for small animal studies is constructed . The technical parameters are as follows : the minimum feeding amount of the fault cutting equipment is 0.001 mm , and the repeated positioning accuracy is 0.01 / 300 mm . The system has the following characteristics : large volume sample imaging ; high spatial resolution ; invasive imaging method ; and the system can obtain accurate structural information of small animals , and provides a new and effective means for the study of small animals . ( 2 ) In this paper , two adult SD rats were used as an example to describe the experimental procedure of tomographic imaging . Based on a tomographic imaging system suitable for small animal studies , SD rats were acquired ( voxel size 20 渭m 脳 20 渭m 脳 20 渭m , image resolution 4 , 600 脳 2 , 580 脳 24 bit , BMP format , total 9475 , 314.68 GB ) and vascular perfusion SD rats . The image resolution was 4 , 917 脳 3 , 446 脳 24 bit , BMP format ( 1646 , 77.9 GB ) . ( 3 ) The technical scheme and the construction technology of constructing the digital anatomic data set were discussed , and the SD rat digital anatomical data set was constructed based on the SD rat tomographic dataset and the vascular perfusion SD rat , and the model of the three - dimensional anatomical structure and the system structure model of SD rats were obtained , which had extensive education and application value . ( 4 ) " Visual Rat Atlas " and " Visual Rat Explorer " respectively show the series of tomographic images , anatomical maps and three - dimensional anatomical structure models of SD rats . The use of the software enables the researchers to analyze and calculate the digital anatomical data sets conveniently , and is a convenient and practical research tool . The research proves that the tomographic imaging system suitable for small animal studies provides an effective means for the study of small animal structure information . The digital anatomical data set constructed on the basis of the system can be used as a framework to integrate more biological information , study the biological information at different levels and levels , and provide an application model for the research of small animals and in biomedical research .
