【摘要】: 目的:通過(guò)建立反流性食管炎(Reflux Esophagitis,RE)動(dòng)物模型及抗反流手術(shù)模型,觀察食管大體形態(tài)和組織學(xué)改變,檢測(cè)食管標(biāo)本中血紅素加氧酶(HO-1)、誘導(dǎo)型一氧化氮合酶(NOS)及熱休克蛋白70(HSP70)水平,分析比較大鼠反流性食管炎中各反流因素的作用,同時(shí)為反流性食管炎尤其胃食管重建手術(shù)后吻合口反流的防治提供實(shí)驗(yàn)基礎(chǔ)以及理論依據(jù)。 方法:選用6周齡健康雄性SD大鼠120只(230~250g),隨機(jī)分為6組,每組20只,建立反流性食管炎SD大鼠模型(單純酸反流、單純膽汁反流、混合反流各20只)、抗反流手術(shù)模型(食管胃套入吻合術(shù)動(dòng)物模型20只)、單純胃食管吻合動(dòng)物模型及假手術(shù)動(dòng)物模型各20只,觀察各模型組動(dòng)物存活情況、食管標(biāo)本肉眼表現(xiàn)及食管標(biāo)本病理組織學(xué)變化(HE染色),并應(yīng)用逆轉(zhuǎn)錄聚合酶鏈反應(yīng)法(RT-PCR技術(shù))檢測(cè)食管中血紅素加氧酶(HO-1)、誘導(dǎo)型一氧化氮合酶(iNOS)及熱休克蛋白(HSP70)的表達(dá)。利用統(tǒng)計(jì)軟件SPSS10.0對(duì)數(shù)據(jù)進(jìn)行統(tǒng)計(jì)學(xué)分析。 結(jié)果 1單純酸反流性食管炎動(dòng)物模型的檢測(cè)結(jié)果:①存活情況:術(shù)后第4、8、12、16周存活率分別為100%、95%、95%、95%。②食管粘膜肉眼表現(xiàn)分級(jí):術(shù)后4周0級(jí)
[Abstract]:Objective: to observe the changes of gross morphology and histology of esophagus by establishing reflux esophagitis reflux model and anti-reflux operation model. The levels of heme oxygenase (HO-1), inducible nitric oxide synthase (NOS) and heat shock protein 70 (HSP70) were measured in esophageal specimens. To analyze and compare the effects of reflux factors in reflux esophagitis in rats, and to provide experimental basis and theoretical basis for the prevention and treatment of anastomotic reflux in rats with reflux esophagitis, especially after gastroesophageal reconstruction. Methods: 120 healthy male SD rats aged 6 weeks were randomly divided into 6 groups with 20 rats in each group to establish the SD rat model of reflux esophagitis (simple acid reflux). There were 20 bile reflux, 20 mixed reflux, 20 anti-reflux operation models (20 gastroesophageal anastomosis animal models, 20 simple gastroesophageal anastomosis animal models and 20 sham operation animal models). The survival of the animals in each model group, the naked appearance of esophageal specimens and the histopathological changes of esophageal specimens were observed by HE staining. The heme oxygenase (HO-1) in esophagus was detected by reverse transcriptase polymerase chain reaction (RT-PCR). The expression of inducible nitric oxide synthase (iNOS) and heat shock protein (HSP70) were analyzed by statistical software SPSS10.0. Results 1 the results of the animal model of simple acid reflux esophagitis were as follows: the survival rate at 1216 weeks after operation was 100%, 95% and 95%, respectively. 95. 2 grade of unaided esophageal mucosa: grade 0 4 weeks after operation
