天然及氧化修飾高密度脂蛋白對HepG2細(xì)胞MSRG轉(zhuǎn)錄影響的初步研究

【摘要】: 為研究代謝綜合征(metabolic syndrome,MS)與動脈粥樣硬化等疾病之間的關(guān)系,本研究室曾通過高糖膳食構(gòu)建了高甘油三酯血癥大鼠模型,其具有代謝綜合征的基本特征。用該大鼠模型肝臟基因的差異表達(dá)序列片段篩選人肝臟cDNA文庫后發(fā)現(xiàn)一條新的基因,國內(nèi)外尚未有關(guān)于該基因的報道。該新基因被暫時命名為代謝綜合征相關(guān)新基因(metabolism syndrome-related novel gene,MSRG)。 為進(jìn)一步了解MSRG與MS的關(guān)系,研究MSRG的功能,我室正在對影響MSRG轉(zhuǎn)錄因素進(jìn)行系列研究。本文以HepG2細(xì)胞為研究對象,,通過設(shè)計(jì)MSRG、β-actin引物及相應(yīng)的Taqman探針,建立了RealtimePCR定量檢測基因轉(zhuǎn)錄水平的方法。對HepG2細(xì)胞內(nèi)MSRG轉(zhuǎn)錄水平進(jìn)行檢測后,發(fā)現(xiàn)MSRG相對于β-actin為低轉(zhuǎn)錄基因。 我們用天然高密度脂蛋白(native high density lipoprotein,N-HDL)及氧化修飾高密度脂蛋白(oxidized high density lipoprotein,OX-HDL)孵育HepG2細(xì)胞,觀察對細(xì)胞MSRG轉(zhuǎn)錄的影響。結(jié)果發(fā)現(xiàn):①在N-HDL 0~100mg/L組及OX-HDL 0~50mg/L組,MSRG轉(zhuǎn)錄隨N-HDL及OX-HDL濃度的增加有下降趨勢。與0mg/L組相比,MSRG在N-HDL 50mg/L、100mg/L組和OX-HDL 50mg/L組轉(zhuǎn)錄水平差異具有統(tǒng)計(jì)學(xué)意義(p<0.05)。當(dāng)濃度高于此范圍,MSRG轉(zhuǎn)錄較最低水平有所升高。與N-HDL 100mg/L組相比,N-HDL 200mg/L組基因轉(zhuǎn)錄有所回升,但無統(tǒng)計(jì)學(xué)差異(p>0.05);與OX-HDL 50mg/L組相比,OX-HDL 100mg/L、200mg/L組MSRG mRNA含量有增高趨勢,且200mg/L組與50mg/L組間基因轉(zhuǎn)錄水平差異具統(tǒng)計(jì)學(xué)意義(p<0.05)。②在N-HDL 0~24h組及OX-HDL 0~12h組,MSRG轉(zhuǎn)錄隨時間延長有下降趨勢。與0h相比,N-HDL 12h、24h、36h組基因轉(zhuǎn)錄降低(p<0.05),24h組MSRG mRNA含量最低;OX-HDL 6h、12h組基因轉(zhuǎn)錄較其0h組降低(p<0.05),12h組MSRG轉(zhuǎn)錄水平最低。當(dāng)時間超過相應(yīng)范圍后,MSRG轉(zhuǎn)錄有回升傾向。與N-HDL 24h組相比,MSRG在N-HDL36h組轉(zhuǎn)錄有所回升,但無統(tǒng)計(jì)學(xué)差異(p>0.05);OX-HDL 24h、36h組MSRG轉(zhuǎn)錄較其12h組有所回升(p<0.05)。③分別用100mg/L N-HDL或OX-HDL孵育HepG2細(xì)胞12h后,MSRG轉(zhuǎn)錄水平均較對照組顯著下降(p<0.05),但二者間無顯著差異。N-HDL、OX-HDL組培養(yǎng)基總膽固醇濃度較孵育前下降,甘油三酯濃度較孵育前上升,但均無統(tǒng)計(jì)學(xué)差異(p<0.05)。細(xì)胞內(nèi)糖原、總膽固醇及甘油三酯濃度在三組間均無顯著性差異。 我們發(fā)現(xiàn):在低濃度及短時間內(nèi),N-HDL及OX-HDL均可降低HepG2細(xì)胞MSRG的轉(zhuǎn)錄水平。N-HDL較OX-HDL作用更為明顯,抑制轉(zhuǎn)錄作用的濃度及時間范圍均寬于OX-HDL。培養(yǎng)基由于加入了高密度脂蛋白,MSRG轉(zhuǎn)錄下降的同時伴有培養(yǎng)基膽固醇、甘油三酯濃度的改變。細(xì)胞內(nèi)糖原、總膽固醇和甘油三酯濃度未發(fā)生顯著變化。N-HDL及OX-HDL影響HepG2細(xì)胞MSRG轉(zhuǎn)錄的機(jī)制尚需進(jìn)一步研究。
[Abstract]:In order to study the relationship between metabolic syndrome ( MS ) and atherosclerosis , a rat model with high triglyceride was constructed by a high - sugar diet , which had the basic characteristics of metabolic syndrome . A new gene was found after screening the human liver cDNA library with the differential expression sequence fragment of the rat model liver gene . The gene was not reported at home and abroad . The new gene was temporarily named as a new gene related to metabolic syndrome ( MSRG ) . In order to further understand the relationship between MSRG and MS , the function of MSRG was studied . In this paper , the transcription level of MSRG was studied by designing MSRG , 尾 - actin primer and corresponding Taq man probe . After detection of MSRG transcript level in HepG2 cells , it was found that MSRG was a low transcription gene with respect to 尾 - actin . Results : 鈶
