【摘要】: 目的探討急性熱應(yīng)激后大鼠中腦導(dǎo)水管周圍灰質(zhì)與腦橋臂旁核Fos與GFAP的表達(dá)情況。方法將大鼠分別置于24℃、34℃、38.5℃或42℃、濕度為60%的恒溫恒濕溫度艙內(nèi)1 h,建立急性熱應(yīng)激大鼠模型,應(yīng)用免疫組織化學(xué)染色檢測中腦導(dǎo)水管周圍灰質(zhì)與腦橋臂旁核Fos蛋白和膠質(zhì)原纖維酸性蛋白(GFAP)表達(dá)情況。結(jié)果1.對照組大鼠中腦內(nèi)的Fos陽性神經(jīng)元很少。34℃組Fos陽性神經(jīng)元有所增加。38.5℃組神經(jīng)元的反應(yīng)達(dá)高峰。42℃組神經(jīng)元反應(yīng)減弱。在實驗組,Fos陽性神經(jīng)元主要分布于中腦導(dǎo)水管周圍灰質(zhì),尤其是腹外側(cè)區(qū)。2.在中腦,GFAP陽性星形膠質(zhì)細(xì)胞的分布區(qū)域與Fos陽性神經(jīng)元類似。24℃組,GFAP陽性星形膠質(zhì)細(xì)胞顯示靜息特點,胞體小,突觸細(xì)長;而34℃組表現(xiàn)為活化型,胞體變大,突起增粗,細(xì)胞數(shù)量增加,GFAP染色加深;38.5℃組最顯著,42℃組則出現(xiàn)胞體形態(tài)不規(guī)則,突起斷裂。3.腦橋臂旁核內(nèi)可見Fos陽性神經(jīng)元,其中在24℃~38.5℃組Fos蛋白的表達(dá)隨溫度增高而增加,38.5℃組為高峰,而在42℃組Fos蛋白表達(dá)又有所減少。4.腦橋內(nèi)GFAP陽性星形膠質(zhì)細(xì)胞的分布特點與Fos陽性神經(jīng)元類似,24℃組,GFAP陽性星形膠質(zhì)細(xì)胞顯示靜息特點,胞體小,突觸細(xì)長;在34℃組表現(xiàn)為活化型,胞體變大,突起增粗的現(xiàn)象,且細(xì)胞數(shù)量增加,GFAP染色加深;38.5℃組最顯著,但在42℃組星形膠質(zhì)細(xì)胞的數(shù)量則有所減少,并出現(xiàn)胞體形態(tài)不規(guī)則、突起斷裂的現(xiàn)象。結(jié)論1.中腦導(dǎo)水管周圍灰質(zhì),尤其是腹外側(cè)區(qū)神經(jīng)元、星形膠質(zhì)細(xì)胞參與熱應(yīng)激反應(yīng)的神經(jīng)免疫調(diào)節(jié),有可能為此調(diào)節(jié)通路的中繼站之一。2.腦橋臂旁核參與熱應(yīng)激反應(yīng)的神經(jīng)免疫調(diào)節(jié),也可能為此調(diào)節(jié)通路的中繼站之一。3.在本實驗中38.5℃是機體保持正常反應(yīng)的極限溫度,在此溫度以下,神經(jīng)元和星形膠質(zhì)細(xì)胞的反應(yīng)隨溫度上升而加強,溫度升至42℃時神經(jīng)元和神經(jīng)膠質(zhì)細(xì)胞受損傷,蛋白表達(dá)又出現(xiàn)減少。
[Abstract]:Objective to investigate the expression of acute heat stress in rat midbrain periaqueductal gray and parabrachial nuclei of Fos and GFAP. Methods the rats were placed in 24 degrees, 34 degrees, 38.5 degrees or 42 degrees Celsius temperature, humidity and constant temperature and humidity cabin in 60% of the 1 h, to establish a rat model of acute heat stress, immune histochemical staining of periaqueductal gray and parabrachial nucleus Fos protein and glial fibrillary acidic protein (GFAP) expression. In the group the experimental group, Fos positive neurons were mainly distributed in the periaqueductal gray matter, especially the ventrolateral.2. in the midbrain, GFAP positive astrocytes and Fos positive neurons in the distribution area of similar.24 C group, GFAP positive astrocytes Glial cells showed characteristics of resting, small cell body, and synaptic slender; 34 C group showed activation, larger cell body, neurite thickening, increase in the number of cells, GFAP staining deepened; the most significant 38.5 C group, 42 C group showed cell morphology was irregular, bulge fracture.3. parabrachial nuclei in the Fos positive neurons, the expression in the 24 to 38.5 DEG C group of Fos protein increased with the temperature of 38.5 DEG C, group peak, and it reduced the expression of Fos and distribution of GFAP positive astrocytes and.4. positive neurons in the pons were similar between the 42 group of Fos protein C, 24 C group, GFAP positive astrocytes glial cells showed characteristics of resting, small cell body, synaptic slender; at 34 DEG C group showed activation, larger cell body, protruding thickening phenomenon, and the increase in the number of cells, GFAP staining deepened; the most significant 38.5 C group, but in 42 the number of C group of astrocytes decreased, and the emergence of The cell body of irregular shape, protruding fracture phenomenon. Conclusion 1. periaqueductal gray matter, especially the ventrolateral neurons, neural immune astrocytes participate in the regulation of heat stress response, may relay station of this regulated pathway of.2. parabrachial nuclei involved in the heat shock response should neuro immune regulation may therefore the relay station of.3. pathway in this experiment is 38.5 DEG C temperature limit of the body to keep normal reaction, below this temperature, the response of neurons and astrocytes with temperature rise and the temperature rise to 42 degrees to strengthen, neurons and glial cells were damaged, and the protein expression decreased.
