【摘要】:目的:對(duì)首次利用狗腎傳代細(xì)胞(MDCK)分離流感病毒進(jìn)行總結(jié),摸索流感病毒細(xì)胞培養(yǎng)分離技術(shù),積累分離的經(jīng)驗(yàn),提高流感監(jiān)測(cè)水平。方法:培養(yǎng)MDCK細(xì)胞,并接種61例經(jīng)咸陽市疾病預(yù)防控制中心流感網(wǎng)絡(luò)實(shí)驗(yàn)室PCR檢測(cè)為陽性的標(biāo)本,收集病變細(xì)胞,經(jīng)紅細(xì)胞凝集實(shí)驗(yàn)、紅細(xì)胞凝集抑制實(shí)驗(yàn)進(jìn)行流感病毒毒株鑒定,同時(shí)在培養(yǎng)液中加入不同濃度谷氨酰胺以研究谷氨酰胺對(duì)MDCK細(xì)胞生長的影響。結(jié)果:從61例實(shí)時(shí)熒光定量PCR檢測(cè)陽性流感病例標(biāo)本中分離出25株流感毒株,HA滴度均≥1∶16,血凝抑制實(shí)驗(yàn)(HI)結(jié)果顯示HI效價(jià)均≥1∶640,其中18株為A(H3N2)亞型株,7株為A(H1N1)亞型株;谷氨酰胺濃度降至0.1%以下時(shí),細(xì)胞生長速率低,細(xì)胞培養(yǎng)至48 h活性即開始下降。結(jié)論:利用MDCK細(xì)胞培養(yǎng)分離流感病毒在咸陽市取得成功,并發(fā)現(xiàn)谷氨酰胺濃度對(duì)MDCK細(xì)胞的活性和細(xì)胞凋亡有著顯著的影響。
[Abstract]:Objective: to summarize the method of isolation of influenza virus by using MDCK for the first time, to explore the technique of cell culture and isolation of influenza virus, and to accumulate the experience of isolation. Methods: MDCK cells were cultured and 61 samples were inoculated with positive PCR from influenza network laboratory of the Center for Disease Control and Prevention of Xianyang City to collect pathological cells. The influenza virus strains were identified by erythrocyte agglutination test and erythrocyte agglutination inhibition test. At the same time, different concentrations of glutamine were added to the culture medium to study the effect of glutamine on the growth of MDCK cells.
