【摘要】: 胚胎著床是人和哺乳動(dòng)物生殖過程中妊娠建立的第一步,是決定妊娠成功與否的關(guān)鍵。成功的植入取決于處于接受態(tài)的子宮內(nèi)膜和具有侵入性的胚胎間的同步協(xié)調(diào)反應(yīng)。在植入開始,母體與胚胎建立了密切的物質(zhì)和信息交流。目前認(rèn)為有多種分子和細(xì)胞水平的調(diào)控分子如內(nèi)分泌、旁分泌、鄰分泌調(diào)節(jié)因子參與胚胎的著床過程,并通過多種細(xì)胞信號途徑從時(shí)間和空間上對胚胎和子宮進(jìn)行精細(xì)調(diào)控。但是子宮內(nèi)膜僅在某一特定時(shí)期允許胚胎著床,這一時(shí)期時(shí)間很短,代表著子宮內(nèi)膜對胚胎具有容受性,稱為“著床窗口期”。胚胎著床過程包括脫去透明帶、定位、黏附、侵入到最后著床等幾個(gè)階段。由于涉及到眾多基因的調(diào)控以及錯(cuò)綜復(fù)雜的調(diào)節(jié)因子網(wǎng)絡(luò)的參與,早期的胚胎發(fā)育和著床的詳細(xì)分子機(jī)制尚不清楚。 幾乎所有生物學(xué)變化都與關(guān)鍵基因的表達(dá)有關(guān)。人們以往一直試圖從某一特定的分子與基因作為突破口,探尋著床發(fā)生機(jī)制,F(xiàn)在大規(guī)模的基因表達(dá)譜分析已廣泛應(yīng)用于生物學(xué)和醫(yī)學(xué)研究領(lǐng)域,其中寡核苷酸芯片、DNA微陣列以及基因表達(dá)系列分析是目前應(yīng)用最廣泛的基因表達(dá)分析方法。但DNA微陣列只能檢測已知基因,且定量分析受到限制,而SAGE技術(shù)能較完整地獲得基因組表達(dá)豐度的數(shù)量信息,能夠大規(guī)模對轉(zhuǎn)錄本進(jìn)行定性和定量分析,可充分了解基因轉(zhuǎn)錄組的全貌,甚至可以發(fā)現(xiàn)新基因,被證明是一種對全基因組基因進(jìn)行表達(dá)分析和尋找差異表達(dá)基因的強(qiáng)有力工具。通過對轉(zhuǎn)錄本特定位置的短寡核苷酸標(biāo)簽(即SAGE標(biāo)簽)的鑒定、分析可以獲取基因表達(dá)的信息。
[Abstract]:Embryo implantation is the first step of the reproductive process and the establishment of pregnancy in mammals, is key to successful pregnancy. Successful implantation depends on the endometrium is in the receiving state and has invaded synchronous reactive embryos. In maternal and embryo implantation, established the close material and information exchange. At present there are many molecules that control the molecular and cellular levels such as endocrine, paracrine and juxtacrine regulator in embryo implantation process, and through a variety of cellular signaling pathways from time and space of the embryo and the uterus for fine regulation. But the endometrium only in a specific period of time to allow embryo implantation during this period a short, representing the endometrial receptivity has on embryo, known as the "implantation window". The process of embryo implantation includes taking off transparent, positioning, adhesion, invasion to the end with bed etc. The molecular mechanism of early embryo development and implantation is not clear due to the regulation of many genes and the involvement of complex regulatory network.
The expression of almost all biological changes and key genes. People in the past has been trying as a breakthrough from a particular gene and molecular mechanism of implantation. Now, explore the large-scale gene expression profile analysis has been widely applied in the fields of biology and medical research, the oligonucleotide microarray, DNA microarray and serial analysis of gene expression is expressed at present the most widely used gene analysis method. But the DNA microarray can detect known genes, and quantitative analysis is limited, and SAGE technology can better obtain the expression of the number of genomic information abundance, can be large-scale on the transcription of the qualitative and quantitative analysis, can fully understand the gene transcription group picture, can even find new genes, is that is a powerful tool for analyzing and identifying differentially expressed genes on the whole genome. Based on this. Specific location of the short oligonucleotide label (SAGE tag) identification, analysis can obtain the information of gene expression.
