水痘—帶狀皰疹病毒糖蛋白E DNA疫苗的免疫原性研究

【摘要】:水痘-帶狀皰疹病毒(VZV)為DNA雙鏈病毒,長度約為125000個(gè)堿基對(duì)。現(xiàn)已發(fā)現(xiàn)VZV基因組有69個(gè)開放讀碼框(ORFs),至少可以編碼6種糖蛋白,相對(duì)分子量從20×10~3到118×10~3,分別為gE、gB、gH、gI、gC和gL。其中糖蛋白E(gE)是第68個(gè)開放讀碼框的編碼產(chǎn)物,初期翻譯產(chǎn)物的相對(duì)分子量為73×10~3,成熟的糖基化產(chǎn)物為88×10~3~98×10~3。糖蛋白E是病毒感染細(xì)胞表面最豐富的糖蛋白,能夠誘導(dǎo)中和抗體和T輔助細(xì)胞反應(yīng),并且含有被CD4~+和CD8~+細(xì)胞識(shí)別的抗原決定簇。 水痘-帶狀皰疹病毒可引起水痘與帶狀皰疹兩種常見疾病,主要感染新生兒、成人和免疫缺陷的患者,后遺神經(jīng)痛是常見的并發(fā)癥。VZV減毒活疫苗(Oka株)已在世界范圍內(nèi)廣泛應(yīng)用多年。雖然該疫苗可以有效的預(yù)防嚴(yán)重的水痘,但是該疫苗仍存在潛伏性和體內(nèi)復(fù)活等問題,所以國外已有學(xué)者開展了關(guān)于VZV新型疫苗的研究。其中所采用的DNA免疫技術(shù)在試驗(yàn)階段已經(jīng)能夠預(yù)防病毒、細(xì)菌和寄生蟲感染,比傳統(tǒng)的以蛋白為基礎(chǔ)的疫苗預(yù)防有更大的優(yōu)勢。 本研究選擇水痘-帶狀皰疹病毒DNA疫苗這一研究領(lǐng)域,構(gòu)建了水痘-帶狀皰疹病毒糖蛋白E DNA疫苗,并用核酸免疫技術(shù)評(píng)價(jià)其免疫原性。
[Abstract]:Varicella zoster virus (VZV) is a double stranded DNA virus, a length of about 125000 base pairs. It has been found that the VZV genome has 69 open reading frames (ORFs), at least 6 encoding glycoprotein, relative molecular weight from 20 * 10~3 to 118 * 10~3, respectively gE, gB, gH. GI, gC and gL. E (gE) is a glycoprotein encoding product of sixty-eighth open reading frames, the initial relative molecular weight of the translation product is 73 * 10~3, glycosylation product maturity is 88 * 10~3 to 98 * 10~3. E protein is the most abundant cell surface glycoprotein of the virus infection can induce neutralizing antibody and T helper cell responses, and contains CD4~+ and identification of CD8~+ cell epitopes.
Varicella zoster virus can cause the two common diseases of varicella and herpes zoster infection, mainly neonatal, adult and immunodeficiency patients with postherpetic neuralgia is a common complication of.VZV live attenuated vaccine (Oka strain) has been widely used in the world for many years. Although the vaccine can effectively prevent severe varicella, but the there are still latent vaccine in vivo and raised other issues, so foreign scholars have done research on new vaccines against VZV. DNA immune technology which had been adopted to prevent the virus in the test phase, bacteria and parasite infection, have more advantages than conventional protein based vaccine prevention.
In the research area of varicella zoster virus DNA vaccine, we constructed varicella zoster virus glycoprotein E DNA vaccine and evaluated its immunogenicity by nucleic acid immunoassay.
