大鼠腦組織嘌呤核苷酸分解代謝關(guān)鍵酶及μ受體基因表達(dá)研究

【摘要】:為了研究大鼠腦組織不同區(qū)域核酸分解代謝關(guān)鍵酶腺苷脫氨酶(ADA)和黃嘌呤氧化酶(XO)活性和基因表達(dá)是否存在差異,進(jìn)而研究給予處理后腦組織不同區(qū)域是否存在腺苷脫氨酶(ADA)和黃嘌呤氧化酶(XO)基因表達(dá)差異提供理論依據(jù),我們?nèi)⌒坌源笫竽X組織,測定其酶活性,并采用RT-PCR及PCR技術(shù)測定其的ADA和XO mRNA的相對含量。本研究結(jié)果顯示:1.大鼠腦組織不同區(qū)域ADA酶活性存在差異,頂葉、顳葉、腦干、海馬ADA酶活性較高;2.大鼠腦組織不同區(qū)域XO酶活性存在差異,頂葉、顳葉、腦干、海馬ADA酶活性較高;3.大鼠腦組織不同區(qū)域ADA mRNA的相對含量存在差異,mRNA含量與酶活性變化基本一致;4.大鼠腦組織不同區(qū)域XO mRNA的相對含量存在差異, mRNA含量與酶活性變化基本一致;5.大鼠腦組織不同區(qū)域μ-R mRNA的相對含量存在差異,中腦、腦干、丘腦、海馬mRNA相對含量較高。以上結(jié)果表明,大鼠組織嘌呤核苷酸分解代謝酶及μ-R的基因表達(dá)存在差異。
[Abstract]:In order to study the difference in activity and gene expression of adenosine deaminase (ADAA) and xanthine oxidase (XO), the key enzymes of nucleic acid catabolism in different regions of rat brain were studied. Then to study whether there are differences in the expression of adenosine deaminase (ADAA) and xanthine oxidase (XO) gene in different regions of brain tissue after treatment, we take the brain tissue of male rats. The enzyme activity was determined. The relative contents of ADA and XO mRNA were determined by RT-PCR and PCR techniques. The results showed that there were differences in ADA enzyme activity in different regions of brain tissue of the rat and parietal lobe. The activity of ADA was higher in temporal lobe, brain stem and hippocampus. 2. The activity of XO enzyme was different in different regions of rat brain. The activity of ADA was higher in parietal lobe, temporal lobe, brain stem and hippocampus. 3. The relative content of ADA mRNA in different regions of brain tissue of rats was similar to that of enzyme activity. 4. The relative content of XO mRNA in different regions of brain tissue of rats was different, and the content of mRNA was basically consistent with the change of enzyme activity. 5. The relative content of 渭 -R mRNA in different regions of rat brain was different. The relative content of mRNA was higher in midbrain, brain stem, thalamus and hippocampus. The gene expression of purine nucleotide catabolism enzyme and 渭-R in rat tissues was different.
