【摘要】:背景與目的: 肥大細(xì)胞激活后脫顆粒釋放類胰蛋白酶、類糜蛋白酶、肝素、組胺及其它化學(xué)介質(zhì)成分,參與許多慢性炎癥反應(yīng)如應(yīng)激誘發(fā)的腸道潰瘍、類風(fēng)濕關(guān)節(jié)炎、間質(zhì)性膀胱炎、硬皮病及克隆氏病等,尤其是在變態(tài)反應(yīng)性疾病中起著至關(guān)重要的作用。變態(tài)反應(yīng)性炎癥包括支氣管哮喘、變應(yīng)性鼻炎、變應(yīng)性皮炎、食物及藥物過(guò)敏等,,發(fā)病率約占全球人口的30%-40%,并有逐年增長(zhǎng)的趨勢(shì)。由于變態(tài)反應(yīng)性炎癥對(duì)人類的危害極大,所以于2000年末被世界衛(wèi)生組織定為三大非傳染性疾病項(xiàng)目之一。近年,越來(lái)越多的證據(jù)證明肥大細(xì)胞激活后釋放的主要介質(zhì)可能是引起變態(tài)反應(yīng)性炎癥發(fā)病的關(guān)鍵機(jī)制。肥大細(xì)胞的這種脫顆粒反應(yīng),既是機(jī)體的一種防御反應(yīng),也是速發(fā)型變態(tài)反應(yīng)和炎癥等病理反應(yīng)的基礎(chǔ),圍繞穩(wěn)定肥大細(xì)胞而產(chǎn)生的療法將會(huì)成為今后一段時(shí)間內(nèi)治療變態(tài)反應(yīng)性疾病的主流。探討肥大細(xì)胞激活途徑及其機(jī)制能給治療變態(tài)反應(yīng)性炎癥及其它與肥大細(xì)胞相關(guān)的疾病的治療提供新思路。 目前對(duì)于肥大細(xì)胞的IgE依賴性激活途徑的研究已進(jìn)行得很深入,其機(jī)制已開(kāi)始明朗;但非IgE依賴性激活途徑的確切作用機(jī)制還不甚明了。本研究所在“長(zhǎng)江學(xué)者”何教授帶領(lǐng)下經(jīng)過(guò)多年努力,提出了肥大細(xì)胞自身放大機(jī)制的假說(shuō),并逐漸完善,這是對(duì)肥大細(xì)胞激活機(jī)制的新發(fā)現(xiàn)。 人被毒蛇咬傷后局部可見(jiàn)明顯組織壞死和炎癥,早期蛇毒與肥大細(xì)胞之間的作用在動(dòng)物上研究過(guò),發(fā)現(xiàn)眼鏡蛇毒等能誘導(dǎo)大鼠橫隔膜、腹膜和幾尼豬肺部肥大細(xì)胞釋放組胺,其激活肥大細(xì)胞的方式是非IgE依賴性的,但蛇毒對(duì)人肥大細(xì)胞的作用尚未見(jiàn)文獻(xiàn)報(bào)道。我們用純化的蛇毒MT、TM-N49與人肺、大腸及扁桃體組織肥大細(xì)胞共同孵育,觀察其對(duì)人肥大細(xì)胞釋放組胺的誘導(dǎo)作用,探討人肥大細(xì)胞非IgE
[Abstract]:Background and Purpose : In recent years , there has been increasing evidence that the main mediators released after activation of mast cells may be a key mechanism for the pathogenesis of allergic inflammation . At present , the study of IgE - dependent activation pathway of mast cells has been deeply studied , and the mechanism has begun to be clear ; however , the mechanism of non - IgE - dependent activation pathway is not very clear . Under the guidance of Professor Ho , Professor of the Yangtze River , the Institute has made a hypothesis about the amplification mechanism of mast cells and perfected it gradually , which is a new discovery of the activation mechanism of mast cells . The effect of venom of venom on mast cells of human mast cells has not been reported in the literature . We have observed the induction of histamine induced by venom of human mast cells by incubating them with purified snake venom MT , TM - N49 and human lung , large intestine and tonsilar tissue mast cells .
