大鼠脊髓全橫斷后相鄰節(jié)段脊髓神經元NT-3的表達

【摘要】:目的 研究大鼠脊髓全橫斷后,NT-3 在損傷部位相鄰節(jié)段脊髓神經元隨時間的表達變化,為進一步探索脊髓損傷后的再生修復機制和神經營養(yǎng)因子治療脊髓損傷提供實驗依據。方法 體重為 250 g 左右成年 SD 大鼠48 只,隨機分為六組。分正常組、假手術組及胸 10 脊髓節(jié)段全橫斷術后 1天、 3 天、 7 天及 14 天組。動物到達存活時間后,應用免疫組織化學SABC 法和圖像分析技術檢測 NT-3 在脊髓損傷相鄰節(jié)段腹、背角的表達和分布。結果 正常組的 NT-3 免疫反應陽性細胞主要分布在腹角的大神經元和少量小神經元及背角的神經元。損傷區(qū)上段的 NT-3 免疫反應陽性細胞數,在脊髓腹、背角的表達,均于術后 1 天組開始明顯增高并達到高峰,術后 3天、7 天組逐漸回落,術后 14 天組接近正常水平。而損傷區(qū)下段的 NT-3 免疫反應陽性細胞數,在脊髓腹、背角的表達均于術后 1 天組開始明顯增高,術后 3 天組達到高峰,于 7 天組開始下降,到 14 天組接近正常水平。結論在脊髓全橫斷術后,脊髓神經元對 NT-3 的需求增加,提示 NT-3 在脊髓修復中起重要作用。
[Abstract]:Objective to investigate the expression of NT-3 in adjacent spinal cord neurons at different time points after spinal cord transection, and to provide experimental evidence for further exploration of regeneration and repair mechanism after spinal cord injury and neurotrophic factor in spinal cord injury. Methods a total of 48 adult SD rats with a weight of 250 g were randomly divided into six groups. It was divided into 1, 3, 7 and 14 days after total transection in the normal group, the sham operation group and the thoracic 10 segment of the spinal cord. After the animals reached their survival time, the expression and distribution of NT-3 in the abdominal and dorsal segments of spinal cord adjacent to spinal cord injury were detected by immunohistochemical SABC and image analysis techniques. Results the NT-3 immunoreactive cells in the normal group were mainly distributed in the large neurons in the ventral angle and a small amount of neurons in the small neurons and the dorsal horn. The number of NT-3 immunoreactive cells in the upper part of the injured area in the ventral and dorsal horn of the spinal cord increased significantly and reached the peak on the 1 day after operation. After 3 days and 7 days, the group gradually decreased, and the group was close to the normal level on the 14 day after operation. The number of NT-3 immunoreactive cells in the spinal cord at the lower part of the injured area increased significantly on the 1 day after operation, and reached the peak on the 3 day after operation. It began to decline in the 7 day group, and it was close to the normal level in the 14 day group. Conclusion after full transection of spinal cord, the demand for NT-3 is increased in spinal cord neurons, suggesting that NT-3 plays an important role in the repair of spinal cord.
