【摘要】：目的觀察帕金森小鼠模型中TLR4通路對Beta-catenin信號的表達影響。方法 C57BL小鼠和TLR4敲除(TLR4knock out,TLR4 KO)小鼠各36只,分別注射生理鹽水和1-甲基-4-苯基-1,2,3,6-四氫吡啶(MPTP)。采用RT-PCR、Westernblot檢測4組之間的Beta-catenin的m RNA和蛋白表達變化,進一步采用免疫熒光的方法檢測黑質(zhì)區(qū)星形膠質(zhì)細(xì)胞中Beta-catenin的共表達變化。結(jié)果 RT-PCR及Western blot結(jié)果顯示,與生理鹽水注射組小鼠相比,C57BL、TLR4KO小鼠在MPTP注射后,mRNA和蛋白表達明顯升高(P0.01);并且,與C57BL小鼠MPTP注射組相比,TLR4KO小鼠MPTP注射組Beta-catenin的mRNA和蛋白表達明顯升高(P0.01)。進一步的免疫熒光結(jié)果顯示,與C57BL小鼠MPTP注射組相比,TLR4KO小鼠MPTP注射組星形膠質(zhì)細(xì)胞(GFAP)中表達的beta-catenin明顯升高。結(jié)論 TLR4通路在亞急性帕金森MPTP模型中,對黑質(zhì)beta-catenin蛋白的表達起重要調(diào)控作用,Toll樣信號與Beta-catenin信號這種互動關(guān)系可能在帕金森發(fā)病機制中起著重要的作用。
