內(nèi)皮抑素通過(guò)調(diào)控PDGFR/ERK信號(hào)通路抑制人成纖維細(xì)胞纖維化的研究(英文)

目的:近年來(lái)發(fā)現(xiàn)內(nèi)皮抑素可以抑制組織纖維化,但是具體機(jī)制不詳。本文旨在研究?jī)?nèi)皮抑素抑制纖維化的作用機(jī)制。創(chuàng)新點(diǎn):首次闡明了內(nèi)皮抑素對(duì)成纖維細(xì)胞PDGFRβ(血小板衍生生長(zhǎng)因子受體β)/ERK(細(xì)胞外調(diào)節(jié)蛋白激酶)信號(hào)通路的影響及其與纖維化的關(guān)系。方法:體外培養(yǎng)人成纖維細(xì)胞,采用血小板衍生生長(zhǎng)因子-BB(PDGF-BB)或轉(zhuǎn)化生長(zhǎng)因子-β1(TGF-β1)建立細(xì)胞纖維化模型,進(jìn)一步應(yīng)用內(nèi)皮抑素處理,通過(guò)檢測(cè)膠原及肌成纖維細(xì)胞表面標(biāo)志等了解內(nèi)皮抑素對(duì)細(xì)胞纖維化和表型轉(zhuǎn)化的抑制作用。進(jìn)而檢測(cè)內(nèi)皮抑素對(duì)成纖維細(xì)胞PDGFRβ/ERK信號(hào)通路的影響,并分析其可能與抗纖維化作用存在的關(guān)聯(lián)。結(jié)論:內(nèi)皮抑素通過(guò)調(diào)控PDGFRβ/ERK信號(hào)通路抑制瘢痕增生,是多靶點(diǎn)的、具有較好的抗纖維化臨床應(yīng)用前景的藥物。
