慢性魚藤酮小鼠模型中鐵與alpha-突觸核蛋白分布的相關(guān)性研究

帕金森。≒arkinson’s disease,PD)是一種常見的神經(jīng)退行性疾病,其主要病理特征包括中腦黑質(zhì)(substantia nigra,SN)多巴胺能神經(jīng)元的選擇性缺失,殘存的神經(jīng)元內(nèi)出現(xiàn)以alpha-突觸核蛋白(alpha-synuclein,α-syn)為主要成分的嗜酸性包涵體即路易小體(Lewy bodies,LBs)。Braak分級學說認為LBs的病理改變是一個從低位腦到高位腦的漸變過程。其典型的臨床表現(xiàn)包括:靜止性震顫、肌強直、運動遲緩、姿勢反射障礙等。而在運動癥狀出現(xiàn)之前,PD病人常出現(xiàn)嗅覺功能障礙、便秘、焦慮、體位性低血壓等非運動癥狀;赑D病人和PD動物模型的大量研究發(fā)現(xiàn),PD病理情況下SN區(qū)鐵水平明顯增高,α-syn異常聚集往往伴有鐵的沉積。鐵能夠促進α-syn聚集,而α-syn作為鐵代謝相關(guān)蛋白也影響鐵的沉積。LBs中的α-syn 90%以上都以磷酸化形式存在,磷酸化修飾后的α-syn可能更易聚集。為闡明鐵沉積與磷酸化alpha-突觸核蛋白(phosphorylated alpha-synuclein,phos-α-syn)之間的分布關(guān)系。本實驗用12月...
