FasL對樹突狀細胞功能的調(diào)控及在真菌感染中的效應研究

樹突狀細胞(dendritic cells,DC)是由Steinman和Cohn在小鼠脾臟中所發(fā)現(xiàn)的一種具有樹枝狀突起的獨特形態(tài)的細胞。DC是目前已知的最強的抗原提呈細胞,它是特異性免疫應答的觸發(fā)者,因為它能夠刺激初始型T細胞(na?ve T cell)活化和增殖,而M?、B細胞等只能觸發(fā)已活化的T細胞或記憶性T細胞發(fā)生免疫應答。而且也有報道表明,活化的DC細胞能產(chǎn)生釋放大量的炎癥因子TNF-α和IL-6等。而大量炎性因子的釋放促進全身炎癥反應綜合征的發(fā)生發(fā)展進一步誘導多器官功能衰竭。故對可調(diào)控DC功能的相關(guān)分子進行研究具有重要意義。同時,細胞凋亡與固有免疫的關(guān)系仍是目前研究的熱點,凋亡相關(guān)分子對固有免疫的調(diào)節(jié)效應也逐漸被人們所關(guān)注。以往研究表明,作為TNFR和TNF家族重要成員的Fas和FasL分子是經(jīng)典的凋亡誘導信號,但近年來的研究表明,Fas和FasL也可以分別傳遞信號,發(fā)揮不同于凋亡誘導的效應。FasL分子與Fas受體結(jié)合后,表達FasL分子的細胞其自身在FasL分子反向信號的作用下發(fā)生相應改變。相關(guān)的體內(nèi)和體外實驗也證明,FasL分子作為共刺激分子參與胸腺細胞成熟和成熟T細胞...

軍醫(yī)大學碩士學位論文) FasL 突變小鼠來源的 BMDC 對 CLR 受體刺激劑的敏感性降低為了研究 DC 的 FasL 反向信號在抗真菌感染中的效應,我們將來源于野生型小鼠sL 胞內(nèi)段突變小鼠 FasL△Intra 來源的 BMDC 用 LPS ,低濃度的 zymd(dectin-1 受活劑)和 mannan(dectin-2 受體激活劑)處理后,ELISA 檢測細胞因子 IL-6,TNF分泌,結(jié)果發(fā)現(xiàn) FasL△Intra 小鼠來源的 BMDC 對 CLR 受體激動劑不敏感,IL-6,TNF分泌明顯低于 WT 小鼠來源的 BMDC(圖 3),這說明 DC 在低劑量 CLR 受體激動激下,F(xiàn)asL 反向信號可能促進 CLR 信號通路的活化。
