　　目的:胸腺基質(zhì)細(xì)胞生成素（Thymic stromal lymphopoietin,TSLP）于20多年前,被鑒定為小鼠胸腺基質(zhì)細(xì)胞系分泌的一種細(xì)胞因子。不久后,相應(yīng)的人類直系同源基因也被發(fā)現(xiàn)。目前,TSLP所介導(dǎo)的信號傳導(dǎo)通路已被廣泛研究,包括上調(diào)多種Th2相關(guān)疾病的細(xì)胞因子,如:哮喘、遺傳過敏性皮炎、超敏反應(yīng),甚至有部分癌癥也與之相關(guān)。所以,TSLP被認(rèn)為是治療相關(guān)疾病的潛在靶標(biāo)^[1]。自1975年雜交瘤技術(shù)問世以來,人們能制備具有良好特異性的單克隆抗體。但由于此類單抗的鼠源性,在用于治療人類疾病時(shí),會產(chǎn)生強(qiáng)烈的人抗鼠抗體反應(yīng)（HAMA）^[2,3],導(dǎo)致臨床應(yīng)用受到限制。目前的抗體藥物領(lǐng)域的研究熱點(diǎn),主要集中于利用基因工程技術(shù),將鼠源抗體進(jìn)行人源化改造^[4-6]。單克隆抗體藥物的發(fā)展先后經(jīng)歷了鼠源抗體、嵌合抗體、人源化抗體和全人源抗體四個(gè)階段^[7]。本實(shí)驗(yàn)平臺前期構(gòu)建大庫容量全人源抗體文庫和噬菌體展示技術(shù),篩選獲得了anti-TSLP-scFv（抗胸腺基質(zhì)細(xì)胞生成素全人源單鏈抗體）,很好的解決了...
