DENV-2感染的HUVEC和Balb/c小鼠受CYM-5442干預(yù)后免疫機(jī)制的研究

目的:利用2型登革病毒(Dengue Virus 2 Type,DENV-2)感染的人臍靜脈內(nèi)皮細(xì)胞(primary human umbilical vein endothelial cells,HUVECs)與免疫細(xì)胞相互作用,并受S1P1受體特異性激動(dòng)劑(CYM-5442)干預(yù)后,對(duì)細(xì)胞因子產(chǎn)生的影響。2、利用CYM-5442處理DENV-2感染的Balb/c小鼠,分析不同器官免疫細(xì)胞的分布特點(diǎn),血清細(xì)胞因子的動(dòng)態(tài)變化,探討其可能發(fā)生的免疫機(jī)制。方法:1.登革Ⅱ型病毒(DENV-2)感染HUVECs后,觀察HUVECs的超微結(jié)構(gòu);2.密度梯度離心PBMC,采用磁珠分選Treg、單核細(xì)胞,利用巨噬細(xì)胞集落刺激因子將單核細(xì)胞誘導(dǎo)分化為巨噬細(xì)胞。3.采用Transwell法,HUVEC分別與Treg、單核細(xì)胞、巨噬細(xì)胞共培養(yǎng),并設(shè)S1P1受體特異性激動(dòng)劑干預(yù)組,檢測(cè)各組HUVECs分泌炎性細(xì)胞因子的水平,檢測(cè)各組Treg分泌抑炎性細(xì)胞因子的水平。4.皮下多點(diǎn)注射Balb/c小鼠,建立DENV感染模型,72h收集外周血,分離血清,接種C6/36細(xì)胞,觀察細(xì)胞病變;qPCR檢測(cè)DENV-2N...
