杭州地區(qū)單核細(xì)胞增生李斯特菌食品分離株分子型別研究

【摘要】:目的研究杭州地區(qū)單核細(xì)胞增生李斯特菌(Listeria monocytogenes)食品分離株的分子分型情況,了解當(dāng)?shù)亓餍兄甑男蛣e特征。方法用脈沖場(chǎng)凝膠電泳(PFGE)和多位點(diǎn)序列分型(MLST)的方法對(duì)單核細(xì)胞增生李斯特菌進(jìn)行分子分型。PFGE結(jié)果進(jìn)行聚類分析,并繪制MLST數(shù)據(jù)的最小生成樹。結(jié)果 6個(gè)血清型組成的133株杭州食品分離株共獲得19個(gè)MLST型別,并發(fā)現(xiàn)1個(gè)新的ST型ST767。ST9和ST121是數(shù)量最多的型別。用AscI和ApaI酶切分別獲得33和45個(gè)PFGE帶型。結(jié)論杭州地區(qū)單核細(xì)胞增生李斯特菌食品分離株分子型別分布廣泛,大部分菌株是可引起人李斯特菌病的Lineage I和Lineage II菌株。食品中單核細(xì)胞增生李斯特菌的污染比較嚴(yán)重,應(yīng)加強(qiáng)監(jiān)測(cè)與管理以防食源性疾病的發(fā)生。
[Abstract]:Objective to study the molecular typing of Listeria monocytogenes (Listeria monocytogenes) food isolates in Hangzhou and to understand the typological characteristics of local epidemic strains. Methods the molecular typing of Listeria monocytogenes was carried out by pulse field gel electrophoresis (PFGE) and multi-site sequence typing (MLST). The results of cluster analysis were carried out, and the minimum generating tree of PFGE data was drawn. Results A total of 19 MLST types were obtained from 133 Hangzhou food isolates composed of 6 serotypes, and 1 new St type ST767.ST9 and ST121 were found to be the most numerous. 33 and 45 PFGE bands were obtained by AscI and ApaI, respectively. Conclusion the molecular types of Listeria monocytogenes food isolates are widely distributed in Hangzhou, and most of the strains are Lineage I and Lineage II strains which can cause human Listeria monocytogenes. The contamination of Listeria monocytogenes in food is serious, so surveillance and management should be strengthened to prevent the occurrence of food-borne diseases.
