蝙蝠蛾擬青霉和中國彎頸霉蛋白質(zhì)氨基酸分析及質(zhì)量評(píng)價(jià)

【摘要】:目的對(duì)蝙蝠蛾擬青霉(Paecilomyces hepiali)和中國彎頸霉(Tolypocladium sinenis)2種蟲生真菌菌絲的蛋白質(zhì)氨基酸進(jìn)行含量分析及質(zhì)量評(píng)價(jià),為2種真菌的其他相關(guān)研究和應(yīng)用提供依據(jù)。方法采用鹽酸水解法水解菌絲粉,用Agilent 1200高效液相色譜儀C18反相柱梯度進(jìn)行分離,上柱前加入鄰苯二甲醛-氯甲酸芴甲酯(OPA-FMOC)自動(dòng)衍生化。參照標(biāo)準(zhǔn)品氨基酸,計(jì)算待測(cè)樣品的氨基酸含量。結(jié)果測(cè)定了17種蛋白水解氨基酸的含量,其中蝙蝠蛾擬青霉中有4種氨基酸(天冬氨酸、纈氨酸、異亮氨酸、亮氨酸)的含量高于中國彎頸霉,中國彎頸霉中有11種氨基酸(谷氨酸、絲氨酸、甘氨酸、蘇氨酸、精氨酸、丙氨酸、酪氨酸、胱氨酸、蛋氨酸、賴氨酸、脯氨酸)的含量高于蝙蝠蛾擬青霉。中國彎頸霉氨基酸總量、必需氨基酸總量、非必需氨基酸總量高于蝙蝠蛾擬青霉,蝙蝠蛾擬青霉支鏈氨基酸含量高于中國彎頸霉。兩者必需氨基酸的模式均符合世界衛(wèi)生組織和聯(lián)合國糧農(nóng)組織(WHO/FAO)提出的必需氨基酸模式譜和參考標(biāo)準(zhǔn)。結(jié)論蝙蝠蛾擬青霉與中國彎頸霉均具有較高的營養(yǎng)價(jià)值,但兩者的蛋白質(zhì)氨基酸含量并不等同。
[Abstract]:Objective to analyze the content and quality of protein and amino acids in the hyphae of Paecilomyces bat (Paecilomyces hepiali) and (Tolypocladium sinenis), so as to provide evidence for other related research and application of the two fungi. Methods the mycelium powder was hydrolyzed by hydrochloric acid hydrolysis and separated by Agilent 1200 high performance liquid chromatography (HPLC) C18 reversed-phase column gradient. Phthalaldehyde-chloromethane methyl ester (OPA-FMOC) was added to the column before the column was added. Referring to the standard amino acids, the amino acid content of the samples to be tested was calculated. Results the contents of 17 kinds of protein hydrolyzed amino acids were determined. Among them, the contents of 4 kinds of amino acids (aspartic acid, valine, isoleucine, Leucine) in Paecilomyces cerevisiae were higher than those in Paecilomyces cerevisiae. The contents of 11 kinds of amino acids (glutamic acid, serine, glycine, threonine, arginine, alanine, tyrosine, cystine, methionine, lysine, proline) in Trichoderma sinensis were higher than those in Paecilomyces bat. The total amino acids, essential amino acids and non-essential amino acids of Paecilomyces cerevisiae were higher than those of Paecilomyces bat, and the content of amino acids in the branch chain of Paecilomyces cerevisiae was higher than that of Paecilomyces cerevisiae. The patterns of essential amino acids are in accordance with the model spectra and reference standards of essential amino acids proposed by the World Health Organization and the Food and Agriculture Organization of the United Nations (WHO/FAO). Conclusion Paecilomyces vulgaris and Paecilomyces cerevisiae have high nutritional value, but the protein amino acid content of Paecilomyces vulgaris and Paecilomyces cerevisiae is not the same.
