基于電泳溫度提升建立大鼠肝快速透明化模型

【摘要】:目的:探索一種基于CLARITY技術(shù)的快速肝組織透明化手段,為該技術(shù)在肝臟上的應(yīng)用提供研究基礎(chǔ)。方法:水凝膠灌注大鼠,取出肝臟待水凝膠凝固后切為1mm薄片。切片隨機(jī)分為兩組,對(duì)照組采用常規(guī)被動(dòng)脂質(zhì)清除法處理,實(shí)驗(yàn)組置于特制電泳裝置中,在外加電場(chǎng)條件下洗滌脂質(zhì),通過提高電泳溫度并量化組織透明度,探索最優(yōu)肝透明化條件。結(jié)果:實(shí)驗(yàn)組在12V電壓下37℃電泳48h時(shí)肝切片相對(duì)透明度達(dá)到最高并保持至96h,隨后相對(duì)透明度開始下降,而在48h電泳清除脂質(zhì)后提高溫度繼續(xù)電泳48h,肝組織切片相對(duì)透明度可進(jìn)一步提高且對(duì)照組脂質(zhì)清除速率明顯低于實(shí)驗(yàn)組。結(jié)論:在12V電壓下肝組織切片37℃電泳48h再輔以52℃電泳48h可以實(shí)現(xiàn)較快速的脂質(zhì)清除。

Methods: the rats were perfused with hydrogel and the liver was taken out and cut into 1mm sheet after hydrogel coagulation. The sections were randomly divided into two groups. The control group was treated with conventional passive lipid removal method. The experimental group was placed in a special electrophoresis device to wash the lipid under the condition of applied electric field, and the tissue transparency was quantified by increasing the electrophoresis temperature.
