智三針對AD模型大鼠腦內(nèi)ChAT及GLUT3表達的影響

【摘要】:目的:觀察"智三針"對AD模型大鼠學(xué)習(xí)記憶能力、大腦皮質(zhì)Ch AT及GLUT3的影響,探討針刺治療AD的機制。方法:采用腹腔注射D-gal聯(lián)合雙側(cè)海馬注射Aβ1-40建立AD模型,按隨機數(shù)字表法分為正常組、模型組、非穴組、智三針組各15只。記錄大鼠Y迷宮測試全天總反應(yīng)時間(TRT)以評估其學(xué)習(xí)記憶水平,采用免疫組化染色法檢測大鼠腦皮質(zhì)內(nèi)Ch AT、GLUT3的表達水平。結(jié)果:造模后,模型組、非穴組、智三針組大鼠的TRT較正常組均延長,4周治療后與模型組、非穴組比較,智三針組大鼠TRT縮短;與正常組比較,模型組大鼠腦皮質(zhì)Ch AT、GLUT3表達水平均顯著降低,與模型組、非穴組比較,智三針組Ch AT、GLUT3表達水平均顯著升高。結(jié)論:"智三針"能提高AD大鼠的學(xué)習(xí)記憶能力,其機制可能與升高大鼠腦內(nèi)Ch AT、GLUT3的表達有關(guān)。
[Abstract]:Aim: to observe the effects of "Zhi San Acupuncture" on learning and memory ability, cerebral cortex Ch AT and GLUT3 in AD model rats, and to explore the mechanism of acupuncture on AD. Methods: the AD model was established by intraperitoneal injection of D-gal and bilateral hippocampal injection of A 尾 1-40. According to the method of random digital table, AD model was divided into normal group, model group, non-acupoint group and three-needle group. The total reaction time (TRT) was recorded to evaluate the learning and memory level of rats. The expression of Ch AT,GLUT3 in the cortex of rats was detected by immunohistochemical staining. Results: the TRT of the model group, the non-point group and the Zhi-San acupuncture group was longer than that of the normal group. After 4 weeks of treatment, compared with the model group and the non-point group, the TRT of the model group was shorter than that of the control group. Compared with the normal group, the expression of Ch AT,GLUT3 in the cerebral cortex of the model group was significantly decreased, and that of the model group and the non-point group was significantly higher than that of the model group and the non-acupoint group. Conclusion: "Zhi San Jing" can improve the ability of learning and memory in AD rats, and its mechanism may be related to the increase of Ch AT,GLUT3 expression in rat brain.
