痛瀉要方對(duì)腹瀉型腸易激綜合征模型大鼠腸道5-HT受體信號(hào)傳導(dǎo)系統(tǒng)的影響

【摘要】:目的:從腸道5-HT受體信號(hào)傳導(dǎo)途徑探討痛瀉要方對(duì)IBS-D模型大鼠內(nèi)臟高敏感性影響的作用機(jī)制。方法:采用隨機(jī)數(shù)字表法,將32只雄性SD大鼠隨機(jī)分為3組:空白對(duì)照組,模型對(duì)照組,痛瀉要方組。采用0.5%醋酸灌腸法來(lái)建立IBS-D內(nèi)臟高敏感大鼠模型。痛瀉要方組予痛瀉要方灌胃(大鼠每日生藥用量9 g/kg),干預(yù)2周。2周后用western blot法測(cè)定小腸和結(jié)腸黏膜組織中5-HTR1、2、3的表達(dá)。結(jié)果:模型對(duì)照組近端小腸、遠(yuǎn)端小腸、近端結(jié)腸和遠(yuǎn)端結(jié)腸的5-HT1R蛋白表達(dá)與空白對(duì)照組比較差異有統(tǒng)計(jì)學(xué)意義(P0.05),痛瀉要方組和模型對(duì)照組相比差異有統(tǒng)計(jì)學(xué)意義(P0.05),與空白對(duì)照組相比差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。模型對(duì)照組近端小腸、遠(yuǎn)端小腸、近端結(jié)腸和遠(yuǎn)端結(jié)腸的5-HT2R蛋白表達(dá)與空白對(duì)照組相比差異有統(tǒng)計(jì)學(xué)意義(P0.05)。痛瀉要方組和模型對(duì)照組相比差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05),與空白對(duì)照組相比差異有統(tǒng)計(jì)學(xué)意義(P0.05)。模型對(duì)照組近端小腸、遠(yuǎn)端小腸、近端結(jié)腸和遠(yuǎn)端結(jié)腸的5-HT3R蛋白表達(dá)與空白對(duì)照組相比差異有統(tǒng)計(jì)學(xué)意義(P0.05)。痛瀉要方組和模型對(duì)照組相比差異有統(tǒng)計(jì)學(xué)意義(P0.05),與空白對(duì)照組相比遠(yuǎn)端結(jié)腸差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:痛瀉要方能夠使IBS-D模型大鼠的內(nèi)臟高敏感性降低,其機(jī)制可能是通過(guò)下調(diào)5-HT1R、5-HT3R的表達(dá)來(lái)降低模型大鼠的內(nèi)臟高敏感性。
[Abstract]:Aim: to investigate the effect of Tongxie decoction on visceral hypersensitivity in IBS-D model rats by means of intestinal 5-HT receptor signal transduction pathway. Methods: 32 male SD rats were randomly divided into three groups: blank control group, model control group and Tongxie Yaofang group. The rat model of IBS-D visceral hypersensitivity was established by 0.5% acetic acid enema. Tongxieyoufang group was treated with Tongxieyoufang (9 g/kg daily crude drug dosage) for 2 weeks. After 2 weeks of intervention, the expression of 5-HTR1O2O3 in small intestine and colon mucosa was determined by western blot method. Results: the expression of 5-HT1R protein in the proximal small intestine, distal small intestine, proximal colon and distal colon in the model control group was significantly higher than that in the blank control group (P0.05). There was significant difference between Tongxieyaofang group and model control group (P0.05), but there was no significant difference compared with blank control group (P0.05). The expression of 5-HT2R protein in the proximal small intestine, distal small intestine, proximal colon and distal colon in the model control group was significantly higher than that in the blank control group (P0.05). There was no significant difference between Tongxieyaofang group and model control group (P0.05), but there was significant difference compared with blank control group (P0.05). The expression of 5-HT3R protein in the proximal small intestine, distal small intestine, proximal colon and distal colon in the model control group was significantly higher than that in the blank control group (P0.05). There was significant difference between Tongxieyangfang group and model control group (P0.05), but there was no significant difference in distal colon compared with blank control group (P0.05). Conclusion: Tongxieyangfang can reduce the visceral hypersensitivity of IBS-D rats, and its mechanism may be that it can decrease the visceral hypersensitivity of IBS-D rats by down-regulating the expression of 5-HT _ 1RN _ (5-HT _ 3R).
