H3N2亞型禽流感病毒及豬流感病毒感染人肺腺癌細胞后增殖的差異性變化

【摘要】:人肺腺癌細胞A549被H3N2亞型禽流感病毒和豬流感病毒感染后,探討不同毒株跨種屬感染人呼吸道組織的可能性和趨勢性。通過觀察病毒感染A549細胞后的細胞病變(CPE)、血凝試驗(HA)和50%組織細胞感染量(TCID50)來比較不同毒株感染細胞后的復(fù)制差異和毒力改變,發(fā)現(xiàn)同一亞型不同來源的流感病毒對A549細胞感染和復(fù)制能力有較大差異,哺乳動物來源的病毒更有感染人呼吸道細胞的趨勢,SW/GX/NS2783/2010有潛在的感染人細胞的能力。感染過程中的細胞因子變化和病毒基因組組成特點是進一步研究的方向,應(yīng)重點關(guān)注具有跨種屬感染趨勢的流感病毒及其特殊分子決定簇的組成和特點。
[Abstract]:Human lung adenocarcinoma cell line A549 was infected with H3N2 subtype avian influenza virus and swine influenza virus. (CPE), hemagglutination test (HA) and 50% histocyte infection (TCID50) were used to compare the difference of replication and virulence of A549 cells infected with virus. It was found that influenza viruses from different sources of the same subtype had significant differences in infection and replication ability of A549 cells. Mammalian viruses were more likely to infect human respiratory tract cells and SW/GX/NS2783/2010 had the potential ability to infect human cells. The changes of cytokines and the characteristics of viral genome composition during infection are the direction of further research. We should focus on the composition and characteristics of influenza viruses and their special molecular determinants which have the trend of cross-species infection.
