【摘要】：目的測定芋螺毒素GI與牛血清白蛋白(BSA)偶聯(lián)物免疫小鼠后產(chǎn)生的抗血清是否具有中和GI毒素作用。方法戊二醛法制備GI-BSA偶聯(lián)物,免疫小鼠,制備GI小鼠抗血清,應(yīng)用抗血清的抗體中和實(shí)驗(yàn),驗(yàn)證其對(duì)GI的解毒作用。結(jié)果 SDS-PAGE蛋白電泳結(jié)果顯示,GI與BSA成功偶聯(lián),GI-BSA在標(biāo)志物相對(duì)分子質(zhì)量(120×10~3)處有兩條新蛋白帶;GI-BSA(99μg/只)每隔2周免疫1次,共免疫4次,第4次免疫后第10天抗血清中和滴度效價(jià)1∶64 000;混合100及200μl小鼠抗血清與≥1 LD_(50)劑量GI,腹腔注射,100μl血清可使1×LD_(50)毒素(16.3μg/kg)組小鼠75%存活,200μl血清可使25.8μg/kg毒素組小鼠75%存活。結(jié)論基于GI-BSA半抗原免疫的小鼠抗血清對(duì)GI毒素具有明顯的解毒作用。
[Abstract]:Objective to determine whether the antiserum produced by the conjugate of conotoxin GI and bovine serum albumin (BSA) (BSA) can neutralize GI toxin in mice. Methods GI-BSA conjugate was prepared by glutaraldehyde method, mice were immunized and anti-serum of GI mice was prepared. The antiserum neutralization test was used to verify the detoxification of GI.
