【摘要】：目的:探討蔞芩止嗽煎能否提高青霉素在慢性阻塞性肺疾病急性加重(AECOPD)痰熱證大鼠模型肺組織中的轉(zhuǎn)運能力。方法:80只Wistar雄性大鼠隨機均分為模型組、氨溴索組、二陳湯組、蔞芩止嗽煎組。用熏吸香煙加氣管內(nèi)注射內(nèi)毒素及鼻腔滴入金黃色葡萄菌,鼓風(fēng)干燥箱中進行風(fēng)熱刺激的方法建立大鼠AECOPD痰熱證模型。用高效液相色譜-質(zhì)譜法檢測各組大鼠不同時段生物標(biāo)本中青霉素的濃度。結(jié)果:15min血清及支氣管肺泡灌洗液中蔞芩止嗽煎組藥物濃度較模型組、氨溴索組及二陳湯組顯著增高(P0.05,P0.01);肺組織中各組比較無統(tǒng)計學(xué)意義,但中位數(shù)比較顯示蔞芩止嗽煎組藥物濃度最高。30min血清、肺組織各組比較無統(tǒng)計學(xué)意義,但在灌洗液中蔞芩止嗽煎組藥物濃度均低于氨溴索組、二陳湯組和模型組(P0.05)。結(jié)論:清熱化痰法指導(dǎo)下的蔞芩止嗽煎能提高青霉素在AECOPD痰熱證大鼠模型肺組織中的藥物濃度,即能增強其在肺組織中的轉(zhuǎn)運能力,為臨床選擇中藥作為COPD急性加重期抗生素治療的輔助用藥提供了實驗依據(jù)。
[Abstract]:Objective: to explore whether the penicillin decoction can improve the transport ability of penicillin in the lung tissue of rats with acute exacerbation of chronic obstructive pulmonary disease (AECOPD) with phlegm heat syndrome. Methods: 80 male Wistar rats were randomly divided into three groups: model group, ambroxol group, Erchen decoction group and trichosanthes Qinzisou decoction group. The rat model of phlegm heat syndrome (AECOPD) was established by cigarette smoking, endotoxin injection into trachea and nasal cavity drip of Grape aureus. The concentration of penicillin was determined by high performance liquid chromatography-mass spectrometry. Results: the drug concentration in serum of 15min and bronchoalveolar lavage fluid was significantly higher than that in model group, ambroxol group and Erchen decoction group (P0.05, P0.01). There was no significant difference in the lung tissue, but the median comparison showed that the drug concentration was the highest in the Luoqingzisou decoction group. There was no significant difference between the 30min serum and the lung tissue group. But in the lavage solution, the drug concentration in the decoction group was lower than that in the ambroxol group, Erchen decoction group and model group (P0.05). Conclusion: the penicillin can increase the concentration of penicillin in the lung tissue of rats with AECOPD phlegm heat syndrome, that is, it can enhance its transport ability in the lung tissue under the guidance of the method of clearing heat and removing phlegm. It provides experimental basis for clinical selection of traditional Chinese medicine as an adjuvant drug for the treatment of acute exacerbation of COPD.
