【摘要】：目的建立模擬卡波西肉瘤發(fā)展過程的移植性卡波西肉瘤的動物模型。方法將36只裸鼠隨機(jī)分為A、B、C、D四個(gè)實(shí)驗(yàn)組,每組各9只。卡波西肉瘤細(xì)胞體外培養(yǎng)并計(jì)數(shù),調(diào)節(jié)細(xì)胞懸液濃度為1×10~8/L、1×10~9/L、5×10~9/L、1×10~(10)/L,各取0.2 ml分別接種于四組實(shí)驗(yàn)組裸鼠背部皮下。觀察各組裸鼠腫瘤長出時(shí)間,腫瘤體積及顏色變化,待瘤體長到36 d時(shí)處死裸鼠,取下腫瘤組織進(jìn)行組織病理學(xué)檢查及免疫組化CD31、CD34、AE1/AE3、S100、HHV-8。結(jié)果卡波西肉瘤細(xì)胞株皮下注射移植成瘤率為100%,腫瘤組織經(jīng)免疫組化CD31、CD34染色陽性證實(shí)為血管來源腫瘤。4個(gè)實(shí)驗(yàn)組中裸鼠腫瘤均由斑點(diǎn)逐漸形成斑塊、結(jié)節(jié),各實(shí)驗(yàn)組小鼠瘤體體積隨時(shí)間延長而逐漸增大,不同細(xì)胞濃度的實(shí)驗(yàn)組小鼠瘤體體積差異無統(tǒng)計(jì)學(xué)意義(P0.05)。第36天不同細(xì)胞濃度的實(shí)驗(yàn)組小鼠瘤體重量差異無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論成功建立了卡波西肉瘤細(xì)胞移植動物模型,且注射卡波西肉瘤細(xì)胞濃度越高,長出腫瘤的時(shí)間越短。
[Abstract]:Objective to establish an animal model of transplanted Kaposi's sarcoma which simulates the development of Kaposi's sarcoma. Methods Thirty-six nude mice were randomly divided into four experimental groups with 9 rats in each group. Kaposi sarcoma cells were cultured and counted in vitro. The cell suspension concentration was adjusted to 1 脳 10 ~ (8) / L ~ (-1) 脳 10 ~ (9) / L ~ (5 脳 10 ~ (9) L ~ (1 脳 10 ~ (10) / L). The tumor growth time, tumor volume and color changes were observed in each group. The nude mice were killed when the tumor was 36 days old, and the tumor tissue was removed for histopathological examination and immunohistochemical CD31,CD34,AE1/AE3,S100,HHV-8.. Results the rate of tumor formation by subcutaneous injection of Kaposi sarcoma cell line was 100. The tumor tissue was confirmed to be vascular origin tumor by immunohistochemical CD31,CD34 staining. The tumor volume of each experimental group increased gradually with the prolongation of time, and there was no significant difference in the tumor volume of the experimental group with different cell concentrations (P0.05). On the 36 th day, there was no significant difference in tumor weight in the experimental group with different cell concentrations (P0.05). Conclusion the animal model of Kaposi sarcoma transplantation was successfully established, and the higher the concentration of Kaposi sarcoma cells was, the shorter the time of tumor growth was.
