空腸彎曲菌致急性周圍神經(jīng)病巴馬香豬模型初步建立

【摘要】:目的:建立空腸彎曲菌(Campylobacter jejuni,Cj)感染致急性周圍神經(jīng)病巴馬香豬模型,初步探討以空腸彎曲菌為感染源引起繼發(fā)免疫相關(guān)疾病的發(fā)病模式,為研究急性運動感覺性軸索性神經(jīng)病(acute motor sensory axonal neuropathy, AMSAN)的發(fā)病機制、其病理生理及空腸彎曲菌的生長特性、感染特性奠定研究模型基礎(chǔ)。 方法:以吉蘭-巴雷綜合征(Guillain-Barre syndrome,GBS)患者分離培養(yǎng)空腸彎曲菌,經(jīng)中國疾病預(yù)防控制中心鑒定為空腸彎曲菌菌株,并經(jīng)美國疾病預(yù)防控制中心鑒定血清型為penner O:19型。用哥倫比亞血培養(yǎng)基于42℃,10%CO2、5%O2、85%N2培養(yǎng)空腸彎曲菌。純化菌種制備1×106菌懸液灌喂巴馬香豬,灌喂前禁食12小時、禁水4小時。分別于灌喂前后分離、鑒定胃腸道菌群。自灌喂當(dāng)日起每日觀察巴馬香豬的臨床癥狀:飲食、大便及肢體活動情況。對出現(xiàn)肢體活動障礙巴馬香豬自出現(xiàn)癥狀7天內(nèi)取材其脊神經(jīng)根、坐骨神經(jīng)及臂叢神經(jīng),進行餓酸染色、鍍銀染色、Luxol-fast-blue染色、免疫組織化學(xué)、透射電子顯微鏡進行神經(jīng)病理觀察。 結(jié)果:空腸彎曲菌自然感染巴馬香豬后,模型動物于1周內(nèi)出現(xiàn)腹脹、腹瀉,2周左右出現(xiàn)肢體活動障礙;神經(jīng)病理檢查可觀察到微管、微絲減少,軸索節(jié)段性萎縮,髓鞘示節(jié)段性回縮,未見脫髓鞘樣變性。神經(jīng)病理學(xué)初步診斷為急性運動、感覺軸索性神經(jīng)病。 結(jié)論:空腸彎曲菌感染巴馬香豬可致急性周圍神經(jīng)病,巴馬香豬可以作為AMSAN研究的模型動物。初步探討了神經(jīng)病理檢查方法。
